Tumor-derived cytokines impair myogenesis and alter the skeletal muscle immune microenvironment

Abstract: Muscle wasting is a decline in skeletal muscle mass and function that is associated with aging, obesity, and a spectrum of pathologies including cancer. Cancer-associated wasting not only reduces quality of life, but also directly impacts cancer mortality, chemotherapeutic efficacy, and surgical outcomes. There is an incomplete understanding of the role of tumor-derived factors in muscle wasting and sparse knowledge of how these factors impact in vivo muscle regeneration. Here, we identify several cytokines/ch… Show more

“…Animal models, including the C26 colon adenocarcinoma mouse model, have also revealed associations between cancer and cardiac atrophy and remodeling (131,132). Although the cancer-derived mediators of cardiac atrophy have not yet been identified, studies suggest that tumor-derived mediators, such as proinflammatory cytokines, promote muscle wasting (133,134). These experimental data and clinical associations strongly suggest deeper connections in the organ crosstalk between cancer and the heart.…”

“…Loss in both body weight and lean mass is a hallmark of CAW. Many studies have implicated heightened inflammatory signaling as a driver of CAW [22][23][24]. We quantitatively assessed concentrations of 31 cytokines in the serum of tumor-bearing and vehicle animals (EveTechnologies) (n = 5 vehicle and 4 tumor).…”

“…It has been well established that cytokines secreted by host immune cells in response to tumors and/or by the tumor itself are associated with muscle wasting and functional limitations [30,31]. High plasma levels of IL-6, TNF-α, INF-γ, and IL-1β have been observed in both tumor-bearing animals and cachectic cancer patients [32,33]. Fatigued women who survive breast cancer have significantly higher serum levels of IL-1ra, sTNF-RII, and neopterin than non-fatigued women [34].…”
