Tumor-Derived Lactate Modifies Antitumor Immune Response: Effect on Myeloid-Derived Suppressor Cells and NK Cells

Abstract: In this study, we explore the hypothesis that enhanced production of lactate by tumor cells, because of high glycolytic activity, results in inhibition of host immune response to tumor cells. Lactate dehydrogenase-A (LDH-A), responsible for conversion of pyruvate to lactate, is highly expressed in tumor cells. Lentiviral vector–mediated LDH-A short hairpin RNA knockdown Pan02 pancreatic cancer cells injected in C57BL/6 mice developed smaller tumors than mice injected with Pan02 cells. A decrease occurred in th… Show more

“…The second edged sword of lactate is its acidic function that together with carbonic acid (43) contributes to the extracellular acidic tumor microenvironment (14). Few reports have demonstrated the devastating impact of lactic acid on several immune cell function in vitro (44) and inhibition of cytolytic NK in vivo (45). Interestingly, the anergic state established at low pH (6-6.5) of tumor-specific CD8(þ) T lymphocytes could be reverted by restoring a physiologic pH (39).…”

“…Oncogene upregulation becomes essential for increased glucose and glutamine metabolism following respiratory impairment (83,165). The metabolic waste products of fermentation can destabilize the morphogenetic field of the tumor microenvironment thus contributing to inflammation, angiogenesis and progression (166)(167)(168). Normal mitochondrial function is necessary for maintaining intracellular calcium homeostasis, which is required for chromosomal integrity and the fidelity of cell division.…”

“…In addition, lactate excreted by tumor cells has been shown to suppress anti-tumor immunity by suppressing natural killer (NK) cells and enhancing myeloidderived suppressor cell (MDSC) functions ( Fig. 2; Husain et al 2013). The lack of effector T-cell infiltration and activation is a hallmark of the PDAC microenvironment (Laheru and Jaffee 2005;Clark et al 2007;Feig et al 2013), although the basis for immune suppression in PDAC is not completely understood.…”
