Tumor necrosis factor-α and interleukin-6 suppress microRNA-1275 transcription in human adipocytes through nuclear factor-κB

Abstract: Obesity is a confirmed risk factor for hyperlipidemia, type-II diabetes, hypertension, and cardiovascular disease. MicroRNAs (miRs) have emerged as an important field of study within energy metabolism and obesity. A previous study demonstrated miR-1275 to be markedly down-regulated during maturation of human preadipocytes. It has been reported that miR-1275 dysregulates expression in several types of cancer and infections. Little is currently known about the regulation of miR-1275 transcription. The aim of the… Show more

“…In addition, miR-1275 was implicated in the regulation of NF-κB and IL-6 ( Plowman and Lagos, 2021 ). The involvement of miR-1275 down-regulation has been established in cancers, infections, and obesity ( Zhou et al, 2017 ). MiR-1275 silences ELK1, an E -twenty-six-domain transcription factor related to adipocyte differentiation, and thus suppresses human visceral pre-adipocyte differentiation ( Pang et al, 2016 ).…”

“…First, miR-1275 was significantly downregulated in the COVID-19 patient cohort (Log 2 FC = −5.48), a miRNA implicated in NF-κB IL-6 regulation. For instance, a separate study noted that the treatment of adipocytes with tumour necrosis factor alpha (TNF-α) and IL-6 reduced miR-1275 expression and that the miR-1275 gene promoter harbours NF-κB-binding sites, allowing transcriptional repression after NF-κB activation [ 66 ]. Therefore, miR-1275 may be a useful biomarker in the identification of an inflammatory state in COVID-19.…”

“…25 Transcription of hsa-miR-1275 could be regulated by the response of tumor necrosis factor (TNF)-a and interleukin (IL)-6 through the NF-kB pathway in obesity. 24 hsa-miR-1275 can inhibit the differentiation of human visceral preadipocytes by suppressing ELK1. 26 The expression of hsa-miR-1275 was downregulated in the serum of pregnant women with fetuses having neural tube defects.…”

“…Among dysregulated miRNAs with significant differences, hsa-miR-1275, with a 17-base pair (bp) intergenic miRNA, is encoded by chromosome 6. 24 A previous study demonstrated that hsa-miR-1275 can function as a tumor suppressor and control hepatocellular carcinoma (HCC) tumor growth by targeting several oncogenic members of the insulin-like growth factor (IGF) axis. 25 Transcription of hsa-miR-1275 could be regulated by the response of tumor necrosis factor (TNF)-a and interleukin (IL)-6 through the NF-kB pathway in obesity.…”
