Turning the Spotlight on Lipids in Non-Apoptotic Cell Death

Abstract: Although apoptosis has long dominated the spotlight, studies in the past two decades have expanded the repertoire of programmed cell death (PCD). Several forms of non-apoptotic regulated cell death have been identified, with important links to organismal homeostasis and different disease pathologies. Necroptosis, ferroptosis, pyroptosis, and NETosis are the major forms of PCD that have attracted attention. Clear biochemical distinctions differentiate these forms of non-apoptotic PCD at the protein and membrane… Show more

“…Lipids have been shown to play key roles in other cellular processes including cell division, [ 8 ] survival, [ 9 ] and cell death, [ 10‐12 ] both as important structural and signaling molecules. A previous study showed that lipid metabolism and oxidation are upregulated during senescence, suggesting that lipid‐induced toxicity might play a role during this process.…”

“…In the recent years lipids have emerged as key regulators of cell fate 24,25 . Increasing amount of evidences have highlighted their functions as triggers, executors or modulators of plasma membrane components that act as platforms for RCD execution 26 . Lipids, displaying different susceptibilities to undergo chemical modifications can execute their functions on cell death subroutines by modulating membrane physicochemical properties 27,28 .…”

“…Thus, the ability to sensitize cancerous cells to programmed cell death through the induction of ferroptosis may provide an effective strategy to mitigate the risk posed by tumor growth and metastasis. Aside from their central role in the regulation of cellular death via ferroptosis, lipids also play key roles in alternative cell death modalities, such as those of necroptosis, pyroptosis, NETosis (as reviewed by [68,69]), and apoptosis (as reviewed [70]).…”
