Two Distinct T Cell Subsets, CD4+ and CD8+CD60+, and Their Cytokines Are Required for In Vitro Induction of Human Ragweed-Specific Memory IgE Responses

Abstract: CD8+CD60+ T cells (80–98% CD45RO+; 20% CD23+) are significantly increased in the blood of serum IgE+ ragweed-sensitized (RS) compared with serum IgE-nonatopic humans (p = 0.001). CD8+CD60+ T cells of the RS patients produced IL-2, IL-4, IL-10, IL-12, IFN-α. and IFN-γ, but not IL-6 or IL-13. When their PBMC were cultured with ragweed Ag (RA), peak IgE responses occurred on day 10; none was induced with non-cross-reacting or without Ag; nonatopic PBMC did not respond to any stimulant. When either CD4+ or CD8+CD6… Show more

“…DMSO (0.1%–1%) (MilliporeSigma, Burlington, MA) was used as the control. As in our earlier studies, levels of IgE in cultures supernatants peaked on day 10 11,12 . On day 10, cells were recovered, counted, viability determined and the levels/extent of phosphorylation of MAPK (P38, ERK and JNK) expressed by leucocyte subsets determined (flow cytometry).…”

“…Distributions of blood lymphocyte (CD4+, CD8+ and CD19+) and monocyte (CD14+) subpopulations were determined as previously described 11,12 . Flow cytometric analysis was performed according to manufacturer's recommendations.…”

“…Several studies have described changes of CD45RO/RA cells in patients with seasonal allergies [1619], however, less is known about changes in central memory and effector memory cell subsets in these patients. In particular, studies describing the changes caused by pollen exposure in T cell subpopulations of patients suffering from allergic rhinitis are required.…”
