Two F‐box/WD‐repeat proteins Pop1 and Pop2 form hetero‐ and homo‐complexes together with cullin‐1 in the fission yeast SCF (Skp1‐Cullin‐1‐F‐box) ubiquitin ligase

Abstract: Background: In the ubiquitin-dependent proteolysis pathway, a ubiquitin ligase (E3) is responsible for substrate selectivity and timing of degradation. A novel E3, SCF (Skp1-Cullin-1/Cdc53-F-box) plays a pivotal role in cell cycle progression. In fission yeast, F-box/WD-repeat protein Pop1 regulates the level of the CDK (cyclin-dependent kinase) inhibitor Rum1 and the S phase regulator Cdc18.

“…6 Phosphorylation of Cdc18 by the cyclin-dependent kinase (CDK) Cdc2 during S phase leads to its ubiquitin-mediated degradation via the SCF Pop1/Pop2 ubiquitin ligase complex. [7][8][9] Cdt1 is degraded during S phase via the Cul4-Ddb1-Cdt2 ubiquitin ligase complex. 10 The overexpression of Cdc18 is sufficient to induce extensive DNA re-replication (with DNA levels ~4-fold higher than in wild type).…”

“…The Cdc4-related Pop1 and Pop2/Sud1 (19 -22) participate in the formation of SCF as either hetero-or homodimers (21)(22)(23). In pop mutants, both the short-lived Cdc2-Cdc13 inhibitor Rum1 and the S-phase regulator Cdc18 were stabilized (20,21). Rum1 is the functional homologue of Sic1 (24), and Cdc18 is the homologue of budding yeast Cdc6 (25).…”

“…As the interaction between Mcs2, Pmh1, and Skp1 did not reflect SCF-dependent regulation, we tested the capacity of Pcu1 [57], the scaffold protein in SCF, to interact with Mcs2 and Pmh1. Three strains were generated expressing both Pcu1 with a myc-tag at the C-terminus (Pcu1-13myc, a kind gift of Tanaka [58]) and TAP-tagged Mcs6, Mcs2, or Pmh1 at their respective endogenous loci.…”
