Two new species of Hypoxylon (Hypoxylaceae) from China based on morphological and DNA sequence data analyses

Abstract: Two new species, Hypoxylon larissae and Hypoxylon jianfengense, are described and illustrated on the basis of a combination of morphological and molecular characters from China. Hypoxylon larissae was collected from southwest China (Yunnan province), while H. jianfengense was collected from southern China (Hainan province). Hypoxylon larissae is characterized by effused-pulvinate stromata, Orange (7) and Luteous (12) pigments extracted in 10% KOH and light brown to dark brown ascospores with straight germ slit… Show more

“…The studied specimens were macromorphologically observed with the aid of a VHX-600E microscope of Keyence Corporation (Osaka, Japan) up to ×200. The microscopic procedure followed Song et al (2022) . Specimen sections were mounted in water, 10% potassium hydroxide (KOH), and Melzer’s reagent (1.5 g potassium iodide, 0.5 g crystalline iodine, and 22 g chloral hydrate dissolved in 20 ml distilled water), and then microscopic examinations were carried out with an Olympus IX73 inverted fluorescence microscope (Tokyo, Japan) at magnifications up to × 1,000.…”

“…The stroma and perithecia were observed, photographed and measured with a VHX-600E 3D microscope from the Keyence Corporation (Osaka, Japan). Fresh material was respectively immersed in water, 10% KOH, and Melzer’s reagent to observe micromorphological structures as determined by Ma et al and Song et al [ 20 , 21 ]. The observations, micrographs, and measurements of asci and ascospores were performed by using an Olympus IX73 inverted fluorescence microscope (Olympus, Tokyo, Japan) and the CellSens Dimensions Software (Olympus, Tokyo, Japan).…”

“…Fresh tissue of stroma was used for DNA extraction and sequence generation following the suggestions by Ma et al and Song et al [ 20 , 21 ]. Sequences of four DNA loci—ITS (internal transcribed spacer regions), nrLSU (nuclear large subunit ribosomal DNA), RPB2 (RNA polymerase II second largest subunit), and β-tubulin (beta-tubulin) were selected for multi-gene phylogenetic analyses [ 2 , 25 ].…”
