Two new species ofKazachstaniathat form ascospores connected by a belt-like intersporal body:Kazachstania zonataandKazachstania gamospora

Abstract: Four strains of ascomycetous yeasts were isolated from samples collected at two locations in southern Japan. The strains formed two warty ascospores that were joined together by an intersporal body appearing as a belt. Phylogenetic analysis of rRNA gene nucleotide sequences indicated that the strains represented two new and closely related species of the genus Kazachstania. Isolates of one of the species were from Miyazaki Prefecture and those of the other species were from the Iriomote Islands. Genetic separa… Show more

“…Purifi cation of PCR products and determination of the sequence of the D1/D2 domain of the LSU rRNA gene were performed as described by Imanishi et al (2007 Reference sequences used for the phylogenetic study, shown in Fig. 1, were obtained from DNA Data Bank of Japan (DDBJ).…”

“…The PCR products were purified with AMPure (Agencourt Bioscience Co.), according to the manufacturer's protocol. Sequence analysis was performed as described previously (Imanishi et al, 2007). Phylogenetic trees were constructed by the neighbour-joining method (Saitou & Nei, 1987) using the CLUSTAL W software (Thompson et al, 1994).…”

“…Purification of PCR products and determination of the sequence of the D1/D2 domain of the 26S rRNA gene were performed as described by Imanishi et al (2007). The sequences determined in this study were deposited in the DDBJ database under the following accession numbers: (=BCC 11765 T =NBRC 105013 T ), DQ404481, AB457175; ST-259 (=BCC 15011=NBRC 105014), AB457176; ST-260 (=BCC 15012=NBRC 105015), AB457177; ST-18 T (=BCC 7718 T =NBRC 105009 T ), DQ400364, AB457171; ST-261 (=BCC 15013=NBRC 105010), AB457172; ST-606 (=BCC 15290=NBRC 105011), AB457173; ST-658 (BCC 15341=NBRC 105012), AB457174.…”
