Two new species of the genus Collodiscula (Xylariaceae) from China

Abstract: Two species similar to Collodiscula were found on bamboo during studies of Xylariaceae in Guizhou Province, China. Phylogenetic analyses of ITS, LSU, RPB2, and β-tubulin loci sequences data indicated that the two taxa are closely related to Collodiscula japonica, the generic type. The new species are introduced in this paper with micrographs and descriptions and compared with C. japonica. Collodiscula leigongshanensis sp. nov., differs from C. japonica by its large (28-35×8-10.5 μm), 3-septate, dark brown asco… Show more

“…The segments of the internal transcribed spacer region (ITS), β-tubulin gene, large-subunit ribosomal RNA gene (LSU), and RNA polymerase II subunit gene (rpb2) were amplified separately by primer pairs, ITS4/ITS5, T11/T22, LR0R/LR5 and RPB2-5f/RPB2-7Cr (Tanaka et al 2009, Hsieh et al 2010, Daranagama et al 2015. PCR amplifications were performed following Li et al (2015) and Su et al (2016). Each reaction mixture comprised a 25 μL total volume, consisting of 19.75 μL of double-distilled water, 2.5 μL of 10× Taq buffer with MgCl2, 0.5 μL of dNTP (10 mM each), 0.5 μL of each primer (10 μM), 0.25 μL Taq DNA polymerase (5 U/μL), and 1.0 μL of DNA template (Li et al 2015).…”

“…The ITS, rpb2, tub2 and α-actin data matrices were combined for subsequent phylogenetic analyses. The sequences (Table 2) used for analyses were retrieved from NCBI-BLAST (https://blast.ncbi.nlm.nih.gov/Blast.cgi) results and some relevant studies (Li et al 2015a, b, Wendt et al 2018, Xie et al 2020, Wu et al 2021. After exclusion of ambiguously aligned regions and long gaps, the final combined data matrix contained 3231 characters (1-444 nucleotides of ITS, 445-1466 nucleotides of rpb2, 1467-2348 nucleotides of tub2 and 2349-3231 nucleotides of α-actin).…”

“…Morphologically, Astrocystis resembles Collodiscula Hino & Katumoto which has superficial carbonaceous stromata, and most of them were discovered from bamboo; meanwhile, the multi-gene analysis indicated that both genera have closer affinities in Xylariaceae (Li et al 2015a, b, Hyde et al 2017, Wu et al 2021. But, species of Collodiscula have larger, one to multi-septate ascospores with a germ slit (except C. leigongshanensis Q.R.…”

“…(1987) , who considered these names morphs of the same species, thus these generic names are synonyms. Three species have been added to Collodiscula ( Li et al 2015a , b ). Given that this generic name has priority and includes four species, the use of Collodiscula is recommended.…”
