Two-Step Potentially Prebiotic Synthesis of α-d-Cytidine-5‘-phosphate from d-Glyceraldehyde-3-phosphate

Abstract: A prebiotically plausible synthesis of ribonucleotides is required to support the "RNA world" hypothesis. 1 The conversion of D-ribose-5-phosphate 1 to the aminooxazoline 2, and thence to R-Dcytidine-5′-phosphate 3, by sequential treatment with cyanamide and cyanoacetylene was demonstrated over 30 years ago (Scheme 1), 2 but no prebiotic synthesis of 1 has been reported. Not only is ribose notoriously difficult to synthesize as anything other than a component of a complex mixture, 3 but it is unstable, 4 and pr… Show more

“…2-aminooxazole 5 is known to be a good C nucleophile at neutral pH, and reacts cleanly with various aldehydes and imines. [27, 4547] For example, when isolated 5 is reacted with 6 in unbuffered water at neutral pH, the reaction proceeds in approximately 90 % yield, with a small diastereoselectivity for the kinetically-preferred rac - 15 over rac - 16 in a 46:43 ratio (Scheme 4). [27] However, the reaction mixture analysed in this pure shift DOSY study shows that rac - 16 predominated over rac - 15 ( rac - 15 / rac - 16 , 1:1.7).…”

“…There is a different potential route for the prebiotic synthesis of pyrimidine nucleotides via arabinose aminooxazoline that first was explored nearly 40 yr ago (Tapiero and Nagyvary 1971) and in recent years has begun to look very persuasive (Ingar et al 2003;Anastasi et al 2007;Powner et al 2009). The earlier studies began with arabinose 3-phosphate, which, like arabinose and other sugars, reacts with cyanamide to give the corresponding aminooxazoline (Fig.…”

“…In principle, RNA can derive from polymerization of activated 5'-nucleotides, or nucleoside-2',3'-cyclic phosphates, and since the latter can easily be produced by cyclization of 2'-or 3'-nucleotides, [8] we initially considered the photoanomerization of all three regio-A C H T U N G T R E N N U N G isomeric mononucleotides. The 5'-nucleotide [9] underwent faster photochemistry, and the b-5'-nucleotide was formed in improved yield, [4,6] but nucleobase loss and oxazolidinone formation were again significant competing reactions. [6] Because oxazolidinone formation is also potentially possible with the 3'-phosphate of 2, we next focused our attention on a-d-cytidine-2'-phosphate 6.…”
