Tyrosine 146 of the Human Na+/Taurocholate Cotransporting Polypeptide (NTCP) Is Essential for Its Hepatitis B Virus (HBV) Receptor Function and HBV Entry into Hepatocytes

Abstract: Na+/taurocholate cotransporting polypeptide (NTCP, gene symbol SLC10A1) is a hepatic bile acid uptake carrier participating in the enterohepatic circulation of bile acids. Apart from its transporter function, NTCP acts as the high-affinity liver-specific receptor for the hepatitis B virus (HBV), which attaches via its preS1-peptide domain of the large surface protein to NTCP, subsequently leading to endocytosis of the virus/NTCP-receptor complex. Although the process of NTCP-dependent HBV infection of hepatocy… Show more

“…Human embryonic kidney Flp-In T-Rex 293 cells (Thermo Fisher Scientific, R78007, hereafter referred to as non-NTCP expressing HEK293 cells) stably expressing the human NTCP protein were generated according to the manufacturer’s guidelines as reported before 62 and are hereafter referred to as HEK-NTCP cells. Functional NTCP expression was confirmed by NBD-TC transport 63 .…”

“…Cells were maintained at 37 °C, 5% CO 2 , and 95% humidity in Dulbecco’s modified Eagle medium (DMEM)/Ham’s F12 medium (Thermo Fisher Scientific, 41966029) supplemented with 10% fetal calf serum (Thermo Fisher Scientific, 10099141), 4 mM L-glutamine (PAA), and antibiotics (100 U/mL penicillin and 100 μg/mL streptomycin, both Anprotec) and 100 μg/mL hygromycin (Carl Roth). For induction of NTCP expression, HEK-NTCP cells were incubated with 1 μg/mL tetracycline (Carl Roth, HP63.1) 62 . Nuclei were stained with Hoechst33342 (Thermo Fisher Scientific, 62249) and fluorescence images were analyzed on a DMI6000 B inverted fluorescent microscope (Leica).…”

“…Human embryonic kidney (HEK293, Thermo Fisher Scientific) cells, stably expressing the human NTCP protein, were generated as described 62 and are further referred to as HEK-15 NTCP cells. Cells were maintained at 37°C, 5% CO2, and 95% humidity in Dulbecco's modified Eagle medium (DMEM)/Ham's F12 medium (Thermo Fisher Scientific) supplemented with 10% fetal calf serum (Sigma-Aldrich), 4 mM L-glutamine (PAA), and antibiotics (100 U/mL penicillin and 100 µg/mL streptomycin, both Anprotec) and 100 µg/mL hygromycin (Carl Roth).…”

“…Posttranslational regulation: To become a functional bile salt transporter and an entry receptor for hepatitis B and D viruses, NTCP is destined to be localized to the hepatocyte plasma membrane. This localization is regulated by several posttranslational modification events (e.g., phosphorylation, glycosylation, S-nitrosylation and ubiquitination; Anwer et al, 2005 ; Kuhlkamp et al, 2005 ; Schonhoff et al, 2011 ; Sargiacomo et al, 2018 ; Zakrzewicz et al, 2022 ).…”
