Ubiquitin E3 Ligase A20 Facilitates Processing Microbial Product in Nasal Epithelial Cells

Abstract: Background: Ubiquitin A20 contributes to the homeostasis. Results: A20 facilitates endosome/lysosome fusion and protein degradation in epithelial cells. Conclusion: A20 is required in degradation of absorbed proteins in epithelial cells. Significance: A20 is a critical molecule in maintaining the homeostasis in the body.

“…Endocytosed antigens are wrapped by plasma membrane to form endosomes, and the latter can fuse with lysosomes; the antigens can then be decomposed by the enzymes in lysosomes. Our data are in line with Li et al () indicating that ubiquitin editing enzyme, A20, facilitates the fusion of endosomes and lysosomes, as observed by An et al (). We found >60% endosomes were fused with lysosomes in the naive states, which was markedly reduced by tryptase in culture.…”

“…21 Similar findings were reported by Li et al, in which Li et al found that deficiency of A20 promoted antigen transport across airway epithelial cells via a transcellular pathway. For example, An et al indicated that the fusion of endosome/lysosome is required in the degradation of microbial products in nasal epithelial cells; the disturbance of the expression of ubiquitin E3 ligase A20 interferes with such a transcellular epithelial barrier function.…”

“…The endocytic proteins can be degraded by the hydrolases in lysosomes (Watts, ). The fusion of endosome/lysosome is important in processing the endocytic proteins in the epithelial cells (An et al, ; Chen et al, ). The factors modulating the fusion of endosome/lysosome are not fully elucidated.…”
