Ubiquitination of G Protein-Coupled Receptors: Functional Implications and Drug Discovery

Abstract: G protein-coupled receptors (GPCRs) comprise the largest and most diverse family of signaling receptors and control a vast array of physiological responses. Modulating the signaling responses of GPCRs therapeutically is important for the treatment of various diseases, and discovering new aspects of GPCR signal regulation is critical for future drug development. Post-translational modifications are integral to the regulation of GPCR function. In addition to phosphorylation, many GPCRs are reversibly modified wi… Show more

“…A different ubiquitination pattern (mono-or polyubiquitination) and how (if several) ubiquitins are attached leads to different fates of the receptor (Alonso and Friedman, 2013). GPCRs have been shown to be affected by ubiquitination and until now processes like internalization, proteasomal degradation, cell signalling, and intracellular trafficking have been reported to be affected (Alonso and Friedman, 2013;Dores and Trejo, 2012). To our knowledge, group I metabotropic glutamate receptors are the only class C receptors that have been shown to be ubiquitinated.…”

“…A recent examination of publicly available gene expression data identified a variety of types of GPCRs (Dores and Trejo, 2012), including protease activated receptor and receptors for various chemokines, adenosine 2B, neuropeptide, metabotropic glutamate, and CaSR that are overexpressed in diverse type of cancer (Rozengurt, 2007) (Figure 1). …”

“…Since then, ubiquitination has been shown to promote lysosomal targeting of many membrane proteins in mammalian cells, including the EGF receptor. 13,35 In the case of many of these proteins, ubiquitin interacts with the ubiquitin-interacting motif (UIM) of Hrs, an endosomal protein often termed ESCRT-0. Hrs then transfers these proteins to Tsg101, an ESCRT-I component that also has a UIM.…”
