Uji Efek Antidiare Ekstrak Rimpang Kunyit Hitam (Curcuma caesia Roxb.) terhadap Mencit (Mus musculus)

Abstract

Diarrhea is a disease characterized by increased frequency of defecation and decreased stool consistency caused by impaired absorption of air and electrolytes in the intestine. Black turmeric is a plant that has properties as a traditional medicine for types of diseases, one of which is diarrhea. The aim of this study was to determine the antidiarrheal effect of black turmeric rhizome extract and the best dose of reducing defecation frequency and increasing stool consistency in diarrheal mice. This research was carried out using 40 mice that were induced by diarrhea using Ricini oleum. Mice were divided into 8 groups, the normal group without treatment, the positive control was given Loperamide HCl, the negative control was given 1% NaCMC, the test control group was given 5 variations of doses of black turmeric rhizome extract 50 mg/KgBW, 150 mg/KgBW, 250 mg/KgBW, 300 mg/KgBW, and 500 mg/KgBW. The results showed that black turmeric rhizome extract could reduce the frequency of defecation and increase stool consistency with a significant value (p<0.05). Black turmeric rhizome extract at a dose of 250 mg/KgBW is the best dose because it can reduce the frequency of defecation and increase stool consistency better than Loperamide HCl.

 Abstrak
 Diare merupakan penyakit yang ditandai dengan peningkatan frekuensi defekasi dan penurunan konsistensi feses disebabkan oleh gangguan absorpsi air dan elektrolit di dalam usus. Kunyit hitam disebut sebagai tanaman yang memiliki khasiat sebagai obat tradisional untuk berbagai jenis penyakit salah satunya diare. Tujuan penelitian ini adalah untuk mengetahui efek antidiare ekstrak rimpang kunyit hitam dan dosis terbaik dalam memberikan efek penurunan frekuensi defekasi dan peningkatan konsistensi feses pada mencit diare. Penelitian ini dilakukan dengan menggunakan 40 ekor mencit yang diinduksi diare menggunakan oleum ricini 0,75 ml. Mencit dibagi menjadi 8 kelompok yaitu kelompok normal tanpa perlakuan, kontrol positif diberi Loperamide HCl, kontrol negatif diberi NaCMC 1%, kelompok kontrol uji diberi 5 variasi dosis ekstrak rimpang kunyit hitam 50 mg/KgBB, 150 mg/KgBB, 250 mg/KgBB, 300 mg/KgBB, dan 500 mg/KgBB. Hasil penelitian menunjukkan bahwa ekstrak ekstrak rimpang kunyit hitam dapat menurunkan frekuensi defekasi dan meningkatkan konsistensi feses dengan nilai signifikasi (p<0,05). Ekstrak rimpang kunyit hitam dosis 250 mg/KgBB merupakan dosis terbaik karena dapat menurunkan frekuensi defekasi dan meningkatkan konsistensi feses dengan hasil lebih baik dari Loperamide HCl.
