Ultracytochemical localization of adenylate cyclase in early human placenta

Abstract: A cytochemical study was carried out on adenylate cyclase (AC) activity in the early human placenta. Samples of placental villi were incubated in a medium containing adenylyl-imidodiphosphate (AMP-PNP) as specific substrate. No AC reaction product was encountered in placenta villi taken at 5 and 7 weeks of pregnancy. AC activity appeared at 9 weeks. At 9 and 10 weeks, AC reaction product was localized on the basal plasma membranes and on apposed plasma membranes of the Langhans cytotrophoblast. At 11 weeks AC … Show more

“…Similar to previous observations on ultracytochemical localization of particulate GC activity (Rambotti et aI., 1987(Rambotti et aI., , 1988Spreca et al, 1989;Rambotti et al, 1990;Spreca et al,, 1991), here we provide morphological evidence that GC reaction product was detected on plasma membranes even if there is some localization of GC reaction product on intracellular membranes. Recently we detected the presence of rANF and pBNP GC capable of stimulating on intracellular membranes in rat adrenal gland (Rambotti et aL, 1993) and in rat neurohypophysis (Rambotti et al, 1994).…”

“…cAMP [38] and membrane-bound stimula tory and inhibitory guanine nucleotide regula tory proteins, Gs and Gi [39], are localized in cytotrophoblasts and syncytiotrophoblasts in early human placenta. Cholera toxin and forskolin stimulated adenylate cyclase and cAMP and hCG production [39,40], suggesting cou pling of Gs to adenylate cyclase.…”
