Ultrasensitive and Rapid Detection of N-Terminal Pro-B-Type Natriuretic Peptide (NT-proBNP) Using Fiber Optic Nanogold-Linked Immunosorbent Assay

Abstract: The N-terminal pro-brain natriuretic peptide (NT-proBNP) is considered an important blood biomarker for heart failure. Herein, we report about a fiber optic nanogold-linked immunosorbent assay (FONLISA) method for the rapid, sensitive, and low-cost detection of NT-proBNP. The method is based on a sandwich immunoassay approach that uses two monoclonal NT-proBNP antibodies, a capture antibody (AbC), and a detection antibody (AbD). AbD is conjugated to a free gold nanoparticle (AuNP) to form the free AuNP@AbD con… Show more

“…In this study, a carboxymethyl dextran (CMD) layer was coated on the RWG substrate surface to minimize the background nonspecific adsorption of UCNP@Ab D onto the sensor surface. Dextran has been shown to reduce nonspecific adsorption of proteins and protein-nanoparticle conjugates onto various sensor surfaces, while the carboxyl groups in CMD provide the binding sites for Ab C conjugation. To eliminate false positive signals due to nonspecific adsorption of UCNP@Ab D onto the RWG substrate surface, we define a threshold value B + 3σ, where B and σ are the average and the standard deviation of the blank, respectively.…”

“…The biosensing system that used simple optical components like LED, PD, analog-to-digital converter, and in-house developed circuit boards has been described previously. 38 Details of the biosensing system are described in the Supporting Information (Section S1), and a schematic is shown in Figure S3. The relative standard deviation (RSD) of the system noise was 5.7 × 10 −5 on average of 300 s. The long-term stability of the optical power level was investigated over a period of 74.1 h, as shown in Figure S4.…”

“…In contrast, by the direct method in an FOPPR biosensor, the binding of one analyte molecule on the AuNP surface will result in an increase of absorptivity of the AuNP, which is several orders of magnitude smaller than the absorptivity of the AuNP itself [ 20 ]. As a result, the FONLISA method improves the sensitivity by at least three orders of magnitude [ 21 , 22 , 23 , 24 , 25 ]. Using this enhancement method, we have pushed the LOD for NNV similar to the level of the gold standard qPCR method, without carrying out amplification steps.…”
