Ultrasensitive Detection of HIV-1 p24 Antigen Using Nanofunctionalized Surfaces in a Capacitive Immunosensor

Abstract: The HIV-1 capsid protein, p24 antigen, is of considerable diagnostic interest because following HIV exposure it is detectable several days earlier than host-generated HIV antibodies (which are the target of almost all current tests used in the field) and can be used to design very sensitive assays without the need for PCR. Here, we present an ultrasensitive capacitive immunosensor that is capable of detecting subattogram per milliliter concentrations of p24 antigen, which to our knowledge is the lowest level o… Show more

“…Capacitive biosensors based on a potential pulse have been used in many applications [1,11,61,62] including microcontact imprinting as seen in Table 3. In this type of capacitive biosensors, a small potential pulse is applied to the working electrode and the capacitance is measured.…”

“…The same strategy was used to develop a capacitive biosensor for sensitive detection of HIV-1 p24 antigen [11]. Following polytyramine electro-polymerization on the gold electrode surface, gold nanoparticles were incorporated onto the electrode and then, anti-HIV-1 p24 monoclonal antibodies were immobilized on top.…”

“…Current HIV-1 diagnostics are largely based on the presence of anti-viral antibody in the blood [28], [29], [30]. Of particular concern is that elevated viral levels during acute infection contribute to as much as 26 times more efficient sexual transmission than that of chronic infection [3], [31].…”
