Ultrastructural changes in oocytes during folliculogenesis in domestic mammals

Abstract: The ultrastructural analysis of oocytes and ovarian follicles has been used to evaluate the effects of assisted reproductive techniques, such as cryopreservation or in vitro oocyte maturation. It also benefits the understanding of such complex mechanisms that occur during folliculogenesis. From the beginning of primordial follicles growth until oocyte maturation in preovulatory follicles oocyte cytoplasmic organelles undergo dynamic alterations that reflect physiological changes and development. This review ai… Show more

“…9,15,16 Alterations in morphology result from complex mechanisms that signal the start or degree of advancement of the atresia process at cellular level, and a wide variety of such alterations have been evaluated to demonstrate their correlation with the atresia signs at histological or ultrastructural level. 23,24 Among histological variables, some that stand out are follicle crenation, the presence of vacuoles, organization of follicular cells around the immature oocyte, and nuclear integrity, all of which were found more frequently in this study. However, the number of morphological alterations increased in follicles treated with OH + NSS.…”

“…Even though the atresia signs can accumulate during folliculogenesis, the existence of incomplete development of the metabolic functions, mainly in the immature oocyte mitochondria, has been reported. 24 This prevents the oocyte from adequately dealing with the damage caused by the oxygen reactive species induced by ethanol at the ultrastructural level during asepsis. Additionally, damage becomes more evident with the acceleration of the atresia process.…”

“…According to Mondadori et al (2010), the location of this organelle into the oocyte varies according to the follicle development and it appears to be more developed in growing follicles than in the early PFs, consistent with the higher metabolism and requirement for protein and lipids of those oocytes (Paulini et al 2014).…”

“…The smallest follicles (primordial and primary) were arranged as groups, forming clusters within the connective tissue, similarly as described for rats (Hirshfield 1989) and chinchillas (Sánchez-Toranzo et al 2014). It is well known that the ovarian cortex contains thousands to millions of primordial follicles that constitute the ovarian reserve from which follicles are engaged for development (Paulini et al 2014).…”

“…As the ovarian follicle grows, the number of lipid droplets associated with endoplasmic reticulum and mitochondria increase, denoting a rise in oocyte metabolism (rev. [34]). During the transformation from primary to secondary follicle, lipid droplets change from small round dark droplets to large gray structures in pigs, which reflects a change in fatty acids composition [35].…”
