Ultraviolet Excitation Photothermal Spectroscopy of Non-Labeled Amino Acids and Visible Light-Induced Signal Enhancement

Abstract: A measurement system for ultraviolet-laser excitation photothermal lens spectroscopy has been designed and constructed for measuring dilute amino acids in liquid solutions. An ultraviolet laser beam is generated as the fourth harmonic of a Ti:sapphire laser in the wavelength range of 212 to 220 nm. Photothermal lens spectra of alanine, phenylalanine, serine, and tryptophan are observed. Photothermal lens spectra of these samples are coincident with their absorption spectra. It is demonstrated that photothermal… Show more

“…For example, reversed-phase mode has versatility for general organic molecules and normal-phase mode could enhance the sensitivity further using organic solvents. Moreover, DIC-TLM can detect biomolecules such as proteins and nucleic acids without fluorescent labeling if its excitation wavelength is tuned to UV region [23,24]. Therefore, the application of DIC-TLM to extended nanochromatography is a giant step for bioanalysis on chip device.…”

“…55) As mentioned above, ultrasound interacts with light, and various measurement methods, devices, and applications based on this principle have been researched, forming a research field called acousto-optics. 30,31,34,39,40,[56][57][58][59][60][61][62][63][64][65][66][67][68][69][70][71][72][73][74][75] Brillouin scattering is caused when light is incident to a medium in which an ultrasonic wave propagates and the light and the ultrasonic wave interact with each other. This phenomenon itself is interesting and is also utilized to measure material properties such as elasticity and so forth.…”

“…To detect non-labeled amino acids in solution by TL measurements, excitation with an UV laser is essential. Recently, our group has successfully detected non-labeled amino acids with a home-built UV-excitation TL detection system using an UV-excitation beam at a wavelength ranging from 212 to 220 nm [8].…”
