Unexpected competitiveness of Methanosaeta populations at elevated acetate concentrations in methanogenic treatment of animal wastewater

Abstract: Acetoclastic methanogenesis is a key metabolic process in anaerobic digestion, a technology with broad applications in biogas production and waste treatment. Acetoclastic methanogenesis is known to be performed by two archaeal genera, Methanosaeta and Methanosarcina. The conventional model posits that Methanosaeta populations are more competitive at low acetate levels (<1 mM) than Methanosarcina and vice versa at higher acetate concentrations. While this model is supported by an extensive body of studies, repo… Show more

“…The limited archaeal diversity may have been due to the lack of niches for microorganisms in coal to inhabit. Other studies have demonstrated the difficulties of amplifying archaeal 16S rRNA gene sequences from coal samples 31 ; however the primer set 515F/806R is known to cover methanogens in wastewater treatment 32 and diverse archaeal communities in coal seams in China 33 . The low archaeal sequence reads may indicate low concentrations of methanogens from the samples, which may result in the observed long lag phase of gas production (Fig.…”

“…Members of these orders were also found in the community in the pilot-scale reactor, but in low relative abundances. Methanothrix species are described to play an important role in the formation and maintenance of a strong granular sludge (MacLeod et al, 1990;McHugh et al, 2005) and it is believed to be dominant at low acetate concentrations (De Vrieze et al, 2012), but it was also recently reported to be able to become dominant at high acetate concentrations (Chen and He, 2015;Chen et al, 2017). Methanobacteriaceae species members could be found in total community, but their presence in the activity community was very low (∼1%).…”

“…While acetate was detected in both day 65 and day 120 samples, OTUs related to the genus Methanosaeta (methanogens that can grow solely on acetate) (12,49) were detected in very low proportions (Ͻ0.5%). Methanosaeta has high affinity for acetate (55) but has often been described to be less competitive at high acetate concentrations (31), although a recent study found strong competitiveness of these methanogens in animal wastewater with an elevated acetate level (59).…”
