Unfolded Protein Response–Related Signature Associates With the Immune Microenvironment and Prognostic Prediction in Osteosarcoma

Abstract: Background: Osteosarcoma is a highly malignant bone tumor commonly occurring in adolescents with a poor 5-year survival rate. The unfolded protein response (UPR) can alleviate the accumulation of misfolded proteins to maintain homeostasis under endoplasmic reticulum stress. The UPR is linked to the occurrence, progression, and drug resistance of tumors. However, the function of UPR-related genes (UPRRGs) in disease progression and prognosis of osteosarcoma remains unclear. Methods: The mRNA expression profiling… Show more

“…Interferonγ has been shown to recruit and activate large amounts of myeloid cells to infiltrate tumor tissue and induce PD-L1 expression to impair tumor immunity effector. 37,38 These results suggested that immune dysregulation in the MSGs model might be a major cause of progression and poor prognosis in 39,40 We evaluated TME in different risk groups and found that patients in the high-risk group had higher immune scores and stromal scores compared with the low-risk group, which might determine a worse prognosis. Previous literature has shown that elevated immune scores and stromal scores in multiple solid tumors correlate closely with poor prognosis, which was consistent with our results.…”

“…These results suggested that immune dysregulation in the MSGs model might be a major cause of progression and poor prognosis in EWS. The tumor microenvironment consists of numerous immune/stromal cells and cytokines, which have a major effect in tumor immune regulation 39,40 . We evaluated TME in different risk groups and found that patients in the high‐risk group had higher immune scores and stromal scores compared with the low‐risk group, which might determine a worse prognosis.…”

“…Recently, bioinformatics analysis combined with machine learning has been widely applied to genomic biodata to detect reliable and robust biomarkers for the diagnosis and treatment of diseases [17,18]. In this study, we utilized bioinformatics analysis to synthesize the role of IRGs based on peripheral blood transcriptomic data from SCI patients.…”

“…Recently, as high-throughput sequencing has evolved, stratifying tumor patients by bioinformatics and machine learning approaches to explore new biomarkers has proven to be reliable and useful ( Zhang et al, 2022a ; Zhang et al, 2022b ; Sun et al, 2022 ). In the current study, we first screened differential expression ofLRGs (DE-LRGs) between EWS and normal tissues.…”
