Unravelling Effects of Anti-aging Drugs on C. elegans using Liposomes

Abstract: Liposome-mediated delivery is a possible means to overcome several shortcomings with C. elegans as a model for identifying and testing drugs that retard aging. These include interactions between drugs and the nematodes’ bacterial food source, and failure of drugs to be taken up into nematode tissues. To explore this, we have tested liposome-mediated delivery of a range of fluorescent dyes and drugs in C. elegans. Liposome encapsulation led to enhanced effects on lifespan, using smaller quantities of … Show more

“…However, feeding metformin to C. elegans increases significant lifespan only in the presence of bacteria. Interestingly, Metformin showed no effect on worms when cultured without bacteria or killed bacteria ( Cabreiro et al, 2013 ; Zhang A. et al, 2023 ).…”

“…The bus-17 or bus-5 mutations, for example, have been used extensively to facilitate drug delivery to C. elegans in other contexts 12 , 124 , 125 . Alternatively, micelle or liposome-forming surfactant or coblock polymers can stabilize the hydrophobic cofactors and facilitate passage of drugs across the cuticle 95 , 126 . In general, due to the thick cuticle of adult C. elegans , incubation at 10 to 100 times the concentration that would be used on isolated cells should be considered.…”

“…[13] In addition, since many drugs with anti-cancer effects, present compounds derived from plants, it is necessary the use of technology to reduce the toxicity that can impair the animals lifespan. [14] Liposomes are a drug delivery system with great biocompatibility, improving biodistribution, promoting controlled drug release and reducing the toxicity of some drugs. [15] Doxil was the first drug approved for cancer treatment using liposomes formulation.…”
