Unravelling the immunological roles of dipeptidyl peptidase 4 (DPP4) activity and/or structure homologue (DASH) proteins

Abstract: SummaryDipeptidyl peptidase (DPP) 4 (CD26, DPP4) is a multi-functional protein involved in T cell activation by co-stimulation via its association with adenosine deaminase (ADA), caveolin-1, CARMA-1, CD45, mannose-6-phosphate/insulin growth factor-II receptor (M6P/IGFII-R) and C-X-C motif receptor 4 (CXC-R4). The proline-specific dipeptidyl peptidase also modulates the bioactivity of several chemokines. However, a number of enzymes displaying either DPP4-like activities or representing structural homologues ha… Show more

“…We hypothesized that the second key target was DPP8, which is 79% similar in protein sequence to DPP9, shares peptide substrate specificity in vitro 17,18 , and, like DPP9, is cytosolic 1923 . Although DPP8 deficient cells released similar basal and Val-boroPro-induced LDH levels as control cells (Fig.…”

“…Intraindividual changes in immune parameters were monitored in patients with type 2 diabetes mellitus in whom gliptin treatment was initiated. Two DPP-IV homologous intracellular dipeptidyl peptidases DPP-8 and DPP-9 are possibly also involved in the functioning of the immune cells (reviewed in [17]). A highly DPP-IV selective inhibitor sitagliptin was therefore chosen to minimize the possibility for off-target effects mediated by the inhibition of DPP-8 and DPP-9 [18].…”

“…Specifically, DPP-4 increases the proliferation of lung fibroblasts and bronchial smooth muscle cells, as well as production of fibronectin from these cells. It is also involved in T-cell activation [37]. As a biomarker, DPP-4 can be measured in the blood using a validated immunoassay [38].…”
