Unsymmetrical Bisquinolines with High Potency against P. falciparum Malaria

Abstract: Quinoline-based scaffolds have been the mainstay of antimalarial drugs, including many artemisinin combination therapies (ACTs), over the history of modern drug development. Although much progress has been made in the search for novel antimalarial scaffolds, it may be that quinolines will remain useful, especially if very potent compounds from this class are discovered. We report here the results of a structure-activity relationship (SAR) study assessing potential unsymmetrical bisquinoline antiplasmodial drug… Show more

“…Third, consider chloroquine, an antimalarial drug, and DM1157 a novel reversed chloroquine compound consisting of a chloroquine backbone with an attached reversal agent (Generous gift from Professor David Peyton and Dr. Katherine Liebman, Portland State University). 2527 Unlike the DOTA and DO3A examples above, chloroquine and DM1157 displayed essentially identical thermodynamic contributions of binding to HSA thermodynamic stability, i.e. Δ H B = ~ 60 kcal/mol, Δ G B = ~ 5 kcal/mol, and Δ T m = ~ 1°C for both compounds.…”

“…Consequently, the antimalarial literature is replete with quinoline and related heterocyclic molecules . Additionally, several hybrid molecules , and bisquinoline molecules are reported to possess potent antiplasmodial activity. The former consists of 4-aminoquinoline pharmacophore in conjunction with other heterocycles, while the latter contains two quinoline rings (total four #Ar) attached with a variable linker.…”

“…Despite the emergence of resistance to 4-aminoquinoline drugs, this pharmacophore has a crucial role inhibiting the heme polymerization and parasite growing. It is believed that the steric hindrance in bulky compounds, such as bisquinolines, would avoid the drug efflux and overcome the resistance [ 35 , 36 , 37 ]. However, hybrids containing the acridine and quinoline moieties are scarcely reported.…”
