Heat shock proteins are highly expressed in various cancers and exert critical functions in tumor progression. However, their expression patterns and functions in lung adenocarcinoma (LUAD) remain largely unknown. We identified that chaperonin-containing T-complex protein-1 subunit 3 (CCT3) was highly expressed in LUAD cells and was positively correlated with LUAD malignancy in the clinical samples. Animal studies showed that silencing CCT3 dramatically inhibited tumor growth and metastasis of LUAD. Proliferation and migration were markedly suppressed in CCT3-deficient LUAD cells. Moreover, the knockdown of CCT3 promoted apoptosis and cell cycle arrest. Mechanistically, the function of glycolysis was significantly inhibited and the total intracellular ATP levels were reduced by at least 25% in CCT3-deficient cells. In addition, the knockdown of CCT3 decreased the protein translation and led to a significant reduction in eukaryotic translation initiation factor 3 (EIF3G) protein, which was identified as a protein that interacts with CCT3. Impaired protein synthesis and cell growth in EIF3G-deficient cells were consistent with those caused by CCT3 knockdown in LUAD cells. Taken together, our study demonstrated in multiple ways that CCT3 is a critical factor for supporting growth and metastasis of LUAD, and for the first time, its roles in maintaining intracellular ATP levels and cytoplasmic translation are reported. Our novel findings provide a potential therapeutic target for lung adenocarcinoma.

Aims Cervical squamous cell carcinoma (SCC) is one of the most frequent malignancies of the female reproductive system. The malignant mechanism of SCC has not been totally clarified. We aimed to discover a list of differentially expressed genes (DEGs) to identify the malignant mechanism of cervical SCC. Method Three expression chips (GSE7803, GSE9750, and GSE64217) were downloaded from gene expression omnibus (GEO) datasets. After standardization, 50 cervical SCC tumor tissues and 33 normal cervical tissues (NCTs) were included for DEGs and clustering analysis. RobustRankAggreg (RRA) algorithm was used to extract the overlapping DEGs. Gene function and signaling pathway analysis was implemented based on Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway databases. Protein–protein interaction (PPI) analysis and prognostic analysis were also carried out to identify the DEGs as prognostic markers for cervical SCC. Results Totally 100 DEGs were obtained from GSE7803, 319 DEGs from GSE9750, and 1639 DEGs from GSE64217. RRA analysis uncovered 17 upregulated DEGs and 25 downregulated DEGs. GO and KEGG analysis showed DEGs were involved in the mediation of extracellular functions, cell–cell interactions, and cell metabolism. PPI network showed a close interaction among the integrated DEGs. Prognostic analysis showed gene secreted phosphoprotein 1 (SPP1) and epiregulin (EREG) genes were independent prognostic predictors of cervical SCC. Conclusion The gene expression profile was changed in cervical SCC tumor tissues compared to NCTs. SPP1 and EREG were postulated as prognostic markers for cervical SCC, which might be potential targets for clinical therapy of cervical SCC.

Genomic analyses identify significant biological processes in DDX21-mediated colorectal cancer cells

Colorectal cancer is the third most common cancer in the US. There has been an incline in the number of young patients with colorectal cancer due to unclear reasons at this point in time. Currently, DEAD-box RNA helicase protein DDX21 is identified as a prognosis marker for early-stage colorectal cancer. However, the mechanism of DDX21 mediated-colorectal cancer is still unknown. Here, our objective is to determine the key molecules and signaling by analyzing the RNA-seq data. The GSE184726 was created by the Illumina NovaSeq 6000 (Homo sapiens). The KEGG and GO analyses indicated Neuroactive ligand−receptor interaction and Ras signaling pathway were the key signaling pathways during the knockdown of DDX21 in colorectal cancer. Moreover, we identified several interactive genes including PTPRC, FN1, ITGAM, RAD51, TRAF6, CCNB1, FOXP3, CCNA2, HIST2H2AC, and HSPA5. Our study may provide new insights into the treatment of colorectal cancer.
