Upregulation of Circular RNA Itchy E3 Ubiquitin Protein Ligase Inhibits Cell Proliferation and Promotes Cell Apoptosis Through Targeting MiR-197 in Prostate Cancer

Abstract: Objective: This study aimed to investigate the effect of circular RNA itchy E3 ubiquitin protein ligase on cell proliferation and apoptosis and to explore its target micro-RNAs in prostate cancer cells. Methods: Circular RNA itchy E3 ubiquitin protein ligase expression in human prostate cancer cells and normal prostate epithelial cells was determined by real time-quantitative polymerase chain reaction assay. Circular RNA itchy E3 ubiquitin protein ligase overexpression plasmids (circular RNA itchy E3 ubiquitin… Show more

“…This circRNA can also down-regulate expression levels of several proteins in the Wnt/b-catenin and PI3K/AKT/mTOR signal transductions in LNCaP and PC-3 cells, as representatives of androgen receptor (AR)-positive and AR-negative cells, respectively (26). miR-197 is another target of circITCH in prostate cancer cells through which it regulates progression of this type of cancer (27).…”

“…miR-17-5p, itself negatively regulates expression of HOXB13, which is known as a tumor suppressor gene being involved in development and progression of PCa. Yuan et al (78) showed the same experimental results about consequences of circ-ITCH down-regulation, although miR-197 was identified as target miRNA for circ-ITCH. Also, miR-17 was detected as target of circ-ITCH in a study by Li et al (79).…”

“…Circular RNA Itchy E3 ubiquitin protein ligase (circ-ITCH) is an example for down-regulated circRNAs in PCa which has been studied in four distinct experiments (77)(78)(79). Wang et al (92) demonstrated that circ-ITCH down-regulation increases PCa cell proliferation and decreases apoptosis in vitro, while its upregulation decreases cell proliferation and in vivo tumor growth.…”

“…Previous studies have reported that miR-197 is significantly overexpressed in the serum of PC patients, which indicates that miR-197 plays an important role in the occurrence and development of PC. However, the molecular mechanism of miR-197 in the bone metastasis of PC has not as yet been reported ( Yuan et al, 2019 ).…”

“…In recent years, abnormal levels of miRNA expression have been found in malignant tumors, which have been found to be closely associated with the proliferation, differentiation, and apoptosis of tumor cells ( Dimakakos et al, 2014 ; Brzeszczyńska et al, 2020 ; Paul et al, 2020 ). Studies have shown that the level of miR-197 expression in the serum of patients with PC is higher than that of benign prostatic hyperplasia patients and healthy males, and the difference is statistically significant ( Yuan et al, 2019 ). These results suggest that miR-197 plays an important role in the occurrence and development of PC.…”
