Upregulation of O‐GlcNAc transferase is involved in the pathogenesis of acute myeloid leukemia

Abstract: Background: Acute myeloid leukemia (AML) is the most common acute leukemia in adults. Patients have a low survival rate and a high recurrence rate, and AML is a highly heterogeneous disease without an effective and specific targeted therapy.Aims: Therefore, it is urgent to explore new AML markers to enable early diagnosis and find drug targets for individualized treatment. Results: Herein, we demonstrate that O-linked-N-acetylglucosamine transferase (OGT) is significantly upregulated in AML tissues compared wit… Show more

“…MLL-AF9 gene defects are formed by reciprocal translocation of t (9;11). Expression of MLL-AF9 fusion genes has been associated with extramedullary tumor infiltration, high recurrence rates, and low survival [ 97 ]. An example is P2X7 (P2X7 belongs to the P2X family of purinergic receptors and is aberrantly expressed in various types of malignant tumors, including leukemia) [ 98 ].…”

“…Currently, O-GlcNAc transferase (OGT) is the only known enzyme in this process affecting cell survival by modulating cell cycle and RAD52 functions. It has been proposed as a novel target in cancer therapies [147,148]. In addition, NEIL3 DNA glycosylase was extensively studied.…”

“…Besides CLL, abnormal O-GlcNAcylation has also been reported in myelodysplastic syndrome (MDS) (Li et al 2021b), acute myeloid leukemia (AML) (He et al 2021), acute lymphoblastic leukemia (ALL) (Zhang et al 2017a), and diffuse large B-cell lymphoma (DLBC) (Pham et al 2016). Interestingly, higher OGT and OGA expression levels were observed in tumor samples and cell lines from hematological malignancies compared with those in other solid tumors (Itkonen et al 2021).…”
