Upregulation of Protein Tyrosine Phosphatase Type IVA Member 3 (PTP4A3/PRL-3) is Associated with Tumor Differentiation and a Poor Prognosis in Human Hepatocellular Carcinoma

Abstract: BackgroundProtein tyrosine phosphatase type IVA member 3 (PTP4A3/PRL-3), a metastasis-associated phosphatase, plays multiple roles in cancer metastasis. We investigated PTP4A3/PRL-3 expression and its correlation with the clinicopathological features and prognosis in hepatocellular carcinoma (HCC).MethodsGene expression profiles of PTP4A3/PRL-3 were obtained in poorly differentiated HCC tissues. The results were validated independently by TaqMan gene expression assays and immunohistochemical analysis.ResultsAc… Show more

“…21 PTP4A3 may increase the potential for invasion by up-regulating MMP (matrix metalloproteinase) in colorectal cancer cell lines 22 and human hepatocellular carcinoma. 23 Taken together it is reasonable to consider PTP4A3 as an oncogene due to its numerous downstream effectors for oncogenesis. It is possible that the function of PTP4A3 is regulated by signals from the microenvironment and not universally identical in different cancer cells in light of the multiple pathways involved with PTP4A3.…”

“…The expression of PRL-3 is frequently upregulated in human tumors, such as breast cancer (21), liver cancer (22), ovarian cancer (23), gastric cancer (24), and colorectal cancer (25). This expression is associated with cancer progression and poor survival.…”

“…This region also contains some cancer related genes such as PTP4A3 and PTK2 genes. The over expression of PTP4A3 has been reported in liver metastases derived from colorectal cancer as well as breast cancer, ovarian cancer, gastric cancer, esophageal carcinoma, and invasive cervical cancer [48]. This gene promotes the cell invasion and activity by stimulating of Rho signaling pathways [49].…”
