Uptake of Al3+ into the N-lobe of human serum transferrin

Abstract: We have studied the binding of Al3+ to human serum apotransferrin (80 kDa) and recombinant N-lobe human apotransferrin (40 kDa) in 0.1 M-sodium bicarbonate solution at a pH meter reading in 2H2O (pH*) of 8.8 using 1H n.m.r. spectroscopy. The results show that for the intact protein, preferential binding of Al3+ to the N-lobe occurs. Molecular modelling combined with an analysis of ring-current-induced shifts suggest that n.m.r. spectroscopy can be used to probe hinge bending processes which accompany metal upt… Show more

“…High fieldshifted resonances have been assigned to protons from residues around Trp 128 , i.e. Leu 122 , and Ile 132 (37,38). The mutation of Ile 132 to Met should lead to the disappearance of the resonance at Ϫ0.603 ppm, which has been previously assigned as ␥CH 2 of Ile 132 .…”

“…3) suggeststhatthenatureofthefirstkineticphaseofthereaction between Yb 3+ and apo-Tf is the same as that of the reaction between Yb 3+ and Fe C -Tf. Therefore the rapid kinetic phase should correspond to the binding of the metal to the N-lobe, although the binding favors the C-lobe thermodynamically as judged by 1 [29,30,32]. The changes in shifts induced by progressive addition of Lu 3+ (and Yb 3+ ) may be due to the conformational change induced by metal ions, a common feature that was observed previously [16,29,30,32].…”

“…The resonance appeared in the high-field region (+0.5 to )1.0 p.p.m., Fig. 5A) was attributed to the methyl groups close to the surface of the aromatic rings of Phe, Trp and Tyr residues due to the ring current effects [29]. Signal b ()0.33 p.p.m) was related to the resonance of the CH 3 group of Leu122, which lies directly above the face of Trp128 [29][30][31].…”
