Uric acid and evolution

Abstract: Uric acid (UA) is the end product of purine metabolism in humans due to the loss of uricase activity by various mutations of its gene during the Miocene epoch, which led to humans having higher UA levels than other mammals. Furthermore, 90% of UA filtered by the kidneys is reabsorbed, instead of being excreted. These facts suggest that evolution and physiology have not treated UA as a harmful waste product, but as something beneficial that has to be kept. This has led various researchers to think about the pos… Show more

“…Analysis of mitochondrial proteins in the livers of SIRT5-overexpressing transgenic mice identified urate oxidase as another target of SIRT5 (103). Urate oxidase catalyzes the conversion of urate to allantoin, the last step of purine catabolism in most mammals (but not humans) (6). In the livers of SIRT5-overexpressing transgenic mice, urate oxidase showed decreased acetylation and increased activity (103).…”

“…In addition, increased urate may mediate blood pressure homeostasis in low-salt environments. Furthermore, higher urate has been suggested to enhance human intelligence or motivational behaviors or promote neuronal integrity and function (4).…”
