Urine protein profiling with surface-enhanced laser-desorption/ionization time-of-flight mass spectrometry

Abstract: SELDI-TOF-MS offers a unique platform for high throughput urine protein profiling; however, standardization of analysis conditions is critical, and both extrinsic and intrinsic factors must be taken into account for accurate data interpretation.

“…However, based on the known molecular weight of cystatin SN (14,312 Da for the mature chain), and the convergence with 2D results, it is most likely that the most discriminant peak corresponds to this protein. In addition, double-charged ions are occasionally generated and detected by SELDI-TOF MS [20]. The peak at m/z 7155 could therefore also correspond to the same compound that would be detected as a doublecharged ion.…”

“…It is found that midstream sample of the second morning urine is optimal and was already used in several studies. [30,31] Urinary proteins have been shown to remain stable long enough to perform reliable proteome analysis. In addition, urine can be stored for several years, even at -20°C, without significant alterations in its proteome.…”

“…tubular epithelial cells) and (b) it may represent the whole kidney allograft. However, urine has variable and changing physicochemical properties (dilution, pH) and cellular components (epithelial cells, leukocytes, red blood cells), which can affect its protein content [11]. Furthermore, stability of proteins under these changing conditions may be impaired [12].…”

“…acute rejection vs. no rejection) can now be analyzed for differences in their respective proteomes. The advantages of SELDI-TOF MS are its user friendliness and high-throughput capabilities [11]. The major disadvantages are a limited sensitivity to detect proteins and a low resolution and mass accuracy of the generated spectra [8,22].…”
