Ursolic Acid fromPlantagomajor, a Selective Inhibitor of Cyclooxygenase-2 Catalyzed Prostaglandin Biosynthesis

Abstract: A hexane extract of Plantago major was investigated by bioactivity-directed fractionation, using an in vitro cyclooxygenase-2 (COX-2) catalyzed prostaglandin biosynthesis inhibition assay, and resulted in the isolation of ursolic acid (1). This triterpenoid showed a significant COX-2 inhibitory effect, directly on the enzyme activity, with an IC50 value of 130 microM and a COX-2/COX-1 selectivity ratio of 0.6. The structural isomer oleanolic acid (2) was found to be less active than 1, with an IC50 value of 29… Show more

“…A number of fatty acids occurring in the human diet, including a-linolenic acid, linoleic acid, docosahexaenoic acid and eicosapentaenoic acid as well as ursolic acid have previously been shown to inhibit COX-2 catalysed prostaglandin biosynthesis and markedly inhibited COX-2 in mice and in in vitro experiments. 33,[44][45][46] In line with this, olive oil and fish oil, which are complex mixtures of fatty acids have been reported to inhibit COX-2 protein expression. 47) Short chain fatty acids are known to modulate different molecular mechanisms of importance in colon cancer cells 48,49) and have been extensively studied for their ability to affect colonic function.…”

“…Oleanolic acid (83) inhibited human leukocyte elastase (Facino et al, 1995;Safayhi and Sailer, 1997). It also inhibited COX-2, and COX-1 in one study (Ringborn et al, 1998) but not in another (Zhang et al, 2004b). Oleanolic acid (83) is a good inhibitor of adenosine deaminase (EC 3.5.4.4) (Koch et al, 1994), one isoform of which is increased in many cancers and immune diseases.…”
