Use of avidin-biotin-peroxidase complex (ABC) in immunoperoxidase techniques: a comparison between ABC and unlabeled antibody (PAP) procedures.

Abstract: The use of avidin-biotin interaction in immunoenzymatic techniques provides a simple and sensitive method to localize antigens in formalin-fixed tissues. Among the 5everal staining procedures available, the ABC method, which involves an application of biotin-labeled secondary antibody followed by the addition of avidin-biotin-peroxidase complex, gives a superior result when compared to the unlabeled antibody method. The availability of biotinbinding sites in the complex is created by the incubation of a relati… Show more

“…For target identification cytospins were stained using Avidin-Biotin Complex (ABC) immunoperoxidase staining [37,38]. Epithelial cells in the pleural effusion cells were identified with a mouse monoclonal antibody (mab) against human pan cytokeratin (clone KL-1, Immunotech, Marseille, France).…”

“…They were then incubated overnight at 41C with anti-MK antibody (human MK goat polyclonal antibody; Santa Cruz Laboratory, Santa Cruz, CA, USA) diluted 1 : 200 in phosphate-buffered saline (PBS) and anti-CD34 antibody (human CD34 mouse polyclonal antibody; Dako Corporation, Carpinteria, CA, USA) diluted 1 : 100 in PBS. The reactions were developed using the avidin -biotin immunoperoxidase technique (ABC method) (Hsu et al, 1981). Immunoreactivity was visualised using the Vectastain Elite ABC kit and a 3,3 0 -diaminobenzidine solution (Vector Laboratories Inc., Burlingame, CA, USA).…”

“…Sections were then incubated with biotinylated rabbit anti-mouse antibody (Dako). Binding sites were visualized using standard avidin-biotin complex method (Hsu et al, 1981). Sections were counterstained with propidium iodide 2.5 µg ml -1 (Sigma).…”
