Use of bioreactor systems in the propagation of forest trees

Abstract: Plant biotechnology can be used to conserve the germplasm of natural forests, and to increase the productivity and sustainability of plantations. Both goals imply working with mature trees, which are often recalcitrant to micropropagation. Conventional in vitro culture uses closed containers and gelled medium with sugar supplementation. Bioreactor culture uses liquid medium and usually incorporates aeration. The increased absorption of nutrients via the liquid medium together with the renewal of the air inside… Show more

“…Temporary or Continuous Immersion Systems (TIS, CIS) were successfully used to proliferate eight ink-resistant C. sativa × C. crenata genotypes [ 145 , 146 ]. The proportion of rootable shoots (tall, vigorous and with an actively growing apex) suitable to undergo the rooting process was increased, and subsequent acclimation produced good plant survival [ 147 ]. Protocols developed for rooting of chestnut include in vitro and ex vitro approaches ( Table 3 ).…”

“…In this context, micropropagation in photoautotrophic conditions in bioreactors can improve rooting and acclimation of recalcitrant tree species, as well as reduce propagation costs and agar-based residues [ 144 , 147 ]. The application of photoautotrophic conditions (high light intensity, CO 2 supplementation, and sugar-free medium) to the multiplication and rooting phase of chestnut has been reported for some genotypes [ 150 ].…”

“…The bioreactor is a modified liquid culture system where the immersion of explant happens periodically. In culture systems with temporary tissue immersion, it is clear that the immersion time is very important and decisive [9] since this factor determines nutrient uptake, which affects the biomass enhancement.…”
