Use of Methanogenic Activity Tests to Characterize Anaerobic Sludges, Screen for Anaerobic Biodegradability and Determine Toxicity Thresholds against Individual Anaerobic Trophic Groups and Species

Abstract: A variety of test procedures have been reported in the literature for determination of the specific methanogenic activity profile of anaerobic sludges and for screening synthetic organic chemicals for anaerobic biodegradability. This paper reviews the methods developed and presents data obtained in the authors' laboratory using test procedures based on pressure transducer measurement of gas pressures in sealed anaerobic vials. Data on the feasibility of freeze-drying granular sludge are also presented as part … Show more

“…The bottles were closed with a septum and the headspace was flushed with N 2 for 5 min to ensure anaerobic conditions. The biogas production was determined by measuring the overpressure in the headspace with time frequency (Colleran et al, 1992). Constant agitation was provided by a shaker at 50 rpm.…”

“…Methanogenic activity tests were performed using the pressure transducer technique 11 (Colleran et al, 1992) to determine the chronic effect of TET on methanogenic pathway. The 12 pressure increase in sealed vials fed with non-gaseous substrates as acetate, propionate, 13 butyrate was monitored.…”

“…Batch specific methanogenic activity (SMA) assays were performed using the seed inoculum and reactor biomass samples from day 673 and day 1243 according to the protocols developed by Colleran et al (1992) and Coates et al (1996), and as described in detail in McKeown et al (2009). At 15 months after the conclusion of the trial, the hydrogenotrophic methanogenic activity of the reactor biomass-which had been stored at 4 1C without receiving any additional substratewas determined.…”
