Use of resistant mutants to study the interaction of triton X-100 with Staphylococcus aureus

Abstract: sitive to phages 52A, 79, and 80) and sensitivity to the cell wall antibiotics penicillin G, vancomycin, bacitracin, and D-cycloserine. Furthermore, RUS3TXR1 retained the Lytphenotype of the parent RUS3. Since mutation was induced by nitrosoguanidine, the mutants would not presumably be isogenic with the wild-type strain.

“…During our characterization of this sensitizing effect, we found that TRX stimulated lipoteichoic acid (LTA) release [9]. This finding was also reported by Raychaudhuri and Chatterjee [11]. LTA is an amphiphilic molecule and anchored to the bacterial cell membrane [4].…”

“…The finding of low lysis induction by daptomycin in strain DU4916 can be considered as a tolerant response to a cell wall antibiotic [18], thus establishing a possible link between this phenomenon in staphylococci [19] and methicillin resistance. In accord with this, lower minimum inhibitory concentrations of vancomycin and D-cycloserine were noted in strain DU4916S than DU4916 [13].…”

“…It is not known how Triton X-100 stimulates S. aureus autolytic activity. Release of the putative autolysin inhibitor lipoteichoic acid may be involved, but it is only a partial explanation [18]. There is evidence for a variety of motifs in autolysin regulation in various bacteria.…”
