Use of Size and a Copolymer Design Feature To Improve the Biodistribution and the Enhanced Permeability and Retention Effect of Doxorubicin-Loaded Mesoporous Silica Nanoparticles in a Murine Xenograft Tumor Model

Abstract: A key challenge for improving the efficacy of passive drug delivery to tumor sites by a nanocarrier is to limit reticuloendothelial system (RES) uptake and to maximize the enhanced permeability and retention (EPR) effect. We demonstrate that size reduction and surface functionalization of mesoporous silica nanoparticles (MSNP) with a polyethyleneimine-polyethylene glycol (PEI-PEG) co-polymer reduces particle opsonization while enhancing the passive delivery of monodispersed, 50 nm doxorubicin-laden MSNP to a h… Show more

“…Very recently, several studies have demonstrated that SN particles not only can serve as promising drug carriers for high tumor targeting efficiency and therapy efficacy, but also for reducing the systematic toxicity [16,17]. However, the biological safety effect is sill being a pendent question of SN particles [18,19].…”

“…Hemolysis assay in vitro was employed to evaluate toxicity of LDMP copolymer (Cerda-Cristerna et al, 2011;Meng et al, 2011). Heparinized rat erythrocytes were separated from rat blood by centrifugation at 3000 r/min for 30 min in a centrifuge and eluted using physiological saline to achromatic color for supernatant solution.…”

“…27,28 SiO 2 -NH 2 -coated SPIO (SPIO@SiO 2 -NH 2 ) nanoparticles prepared by alkaline hydrolysis of APTES have been reported by the present authors 29 and other groups. [30][31][32] In a previous study, SPIO@SiO 2 -NH 2 nanoparticles were applied to rabbit mesenchymal stem cell (MSC) labeling. 29 It was shown that SPIO@ SiO 2 -NH 2 nanoparticles had a higher labeling efficiency than SiO 2 -coated SPIO (SPIO@SiO 2 ) nanoparticles.…”
