Use of solid phase extraction for the isolation and clean-up of a derivatised furazolidone metabolite from animal tissues

Abstract: A method is presented for the determination of protein-bound residues of furazolidone in animal tissue. The use of furazolidone in food-producing animals has been banned in the EU. Illegal use of furazolidone can be monitored most effectively by testing for bound residues containing the 3-amino-2-oxazolidone (AOZ) moiety which, unlike the parent drug, is stable and can be detected for prolonged periods after cessation of treatment. This paper reports the development of an extraction and clean-up procedure for … Show more

“…Tissue bound nitrofuran metabolites are small molecular species which are usually derivatised using ortho-nitrobenzaldehyde (o-NBA) in order to increase molecular mass and improve the sensitivity of detection (Conneely et al, 2002). Prior to derivatisation, the release of bound metabolites from tissue is carried out under mildly acidic conditions (Figure 2b).…”

“…Szilagyi and de la Calle (2006) detected semicarbazide in egg tissue using solid phase extraction in conjunction with LC-MS/MS . Conneely et al (2002) on the other hand, implemented additional clean up measures for the detection of AOZ in pig liver. In this study, a protease digestion step was introduced to overcome matrix effects and allowed the simple application of filtered supernatants to SPE columns.…”

“…As nitrofuran metabolites are covalently bound to proteins and are released from the tissues under slightly acidic conditions, they are derivatised with onitrobenzaldehyde (o-NBA) to form nitrophenyl (NP) derivatives prior to detection [16].…”

“…These methods have been widely used in the extraction of metabolites from animal and plant tissues (Shryock et al, 1986;Kopka et al, 1995;Chen et al, 1998;Adams et al, 1999;Bouchereau et al, 2000;Conneely et al, 2002) and microorganisms (Cordeiro and Freire, 1996;Weuster-Botz, 1997;Hajjaj et al, 1998;Schaefer et al, 1999;Vaseghi et al, 1999;Letisse and Lindley, 2000;Martins et al, 2001;Buchholz et al, 2002;Buziol et al, 2002;Maharjan and Ferenci, 2003). Acidic extraction is commonly used for the extraction of nucleotides and water-soluble metabolites (Vaseghi et al, 1999;Martins et al, 2001;Buchholz et al, 2002), but for metabolome studies it is evident that not all metabolites are stable at the extreme pH, and destruction of some nucleotides has been reported during acidic extractions (Hajjaj et al, 1998).…”
