USH2A-Related Retinitis Pigmentosa: Staging of Disease Severity and Morpho-Functional Studies

Abstract: Usher syndrome type 2A (USH2A) is a genetic disease characterized by bilateral neuro-sensory hypoacusia and retinitis pigmentosa (RP). While several methods, including electroretinogram (ERG), describe retinal function in USH2A patients, structural alterations can be assessed by optical coherence tomography (OCT). According to a recent collaborative study, RP can be staged considering visual acuity, visual field area and ellipsoid zone (EZ) width. The aim of this study was to retrospectively determine RP stage… Show more

“…Moreover, structural OCT and OCT-A are amongst the most effective non-invasive imaging tools for several retinal diseases. As a matter of fact, in some cases, these techniques are even considered to be the first-line diagnostic approaches over the older dye imaging methods [39][40][41].…”

“…Clear vision requires the structural and physiological integrity of all components of the visual system (eyes, brain, and its pathways), and any anomaly results in visual dysfunction. Several factors contribute to the development of visual dysfunction, such as genetic mutations, the environment, lifestyle, age, and malnutrition [ 356 , 357 , 358 , 359 ]. Individuals with visual dysfunction comprise all ages, with a higher prevalence in children and the elderly.…”

“…Sub-groups of patients underwent a more detailed comprehensive electrofunctional study. A total of 12 out of 17 patients performed 30 Hz submicrovolt flicker ERG, (Retimax Advanced Plus, CSO, Scandicci, Italy)) with the assessment of response variability and signal-to-noise ratio (S/N), as already described in a previous article by Falsini et al [10].…”

“…This RP stage score system (RP-SSS) appeared to be readily applicable to different subtypes of RP. In particular, our group [10] recently applied the RP-SSS to the clinical and morphological features of USH2A related retinal degeneration. The USH2A severity score was reliably correlated with patient age as well as with several morphologic and functional parameters of retinal disease [10].…”
