The evolution of the reproductive process has resulted in the development of elaborate adaptive mechanisms to ensure the survival of the offspring. In viviparous animals such adaptations include the development of complex and diverse forms of implantation and placentation in order to support the attachment and development of embryos in utero. The implantation process exhibits remarkable diversity among species, most prominently in the extent of trophoblastic invasion into the uterus. Yet the general aim of implantation is accomplished in all species: to attach the embryo to the uterine wall and to establish an intimate union between maternal and fetal tissues so that an exchange of nutrients and waste products can occur. The details of the earliest interactions between the blastocyst and endometrium have been the focus of extensive study. It is our purpose to summarize some of the more important developments in this field since the publication of Finn's fine review of the subject in the second edition of this book (Finn, 1977). In this chapter we have chosen to discuss four aspects of the implantation process: (1) adhesion of the trophoblast to the uterine epithelium; (2) increased vascular permeability at implantation sites; (3) the decidual cell reaction; and (4) loss of epithelial cells surrounding the implanting blastocyst. Our discussion is based primarily on the results of investigations using common laboratory animals, namely, the rat, mouse, and rabbit. The synthesis of a soluble protein (referred to as 'decidualization-associated protein', DAP), has been examined in uterine and placental tissues of rats during pregnancy by means of polyacrylamide gel electrophoretic analysis of [3H]leucine-labelled soluble proteins. No synthesis of the protein was detected in non-implantation regions of the uterus. In implantation site tissue, no synthesis was detected on Days 6 or 7 of pregnancy. Only slight synthesis was present in the endometrium on Day 8, but synthesis rose rapidly from Days 9 to 12 in both the endometrium and myometrium although differences in the rates of increase were observed. Synthesis fell from Day 12 to 14 in both tissues. Synthesis by the myometrium was entirely localized in the mesometrial region, which contains the metrial gland. After Day 12, when the endometrium is represented by the chorioallantoic placenta, synthesis was examined in the labyrinthine and the decidua basalis/basal zone placenta tissues. No synthesis of 'DAP' was detected in the labyrinthine placenta from Day 16 of pregnancy. Synthesis observed in the decidua basalis/basal zone placenta fell dramatically from Day 14 to 20. The pattern of synthesis of 'DAP' during pregnancy suggests a role in the establishment of the chorioallantoic placenta and metrial gland in the rat.

Summary. Serum, oviducal and uterine luminal fluid proteins from rats at three different physiological stages of reproduction were analysed by polyacrylamide gel electrophoresis. Protein synthesis and secretion were also studied by short-term incubation of tissue pieces in vitro with radioactive leucine. In ovariectomized animals and 5-day pregnant animals, uterine weight and the protein content of the luminal fluid was significantly lower than in pro-oestrus animals. These differences were much less marked in the oviduct. The protein patterns of oviducal fluid and uterine fluid were significantly different from those of blood serum. However, certain proteins, such as those of 51, 56, 68 and 80 K are clearly derived from blood serum. The protein profile of oviducal fluid and extracts of oviducal tissue showed no obvious differences at the three physiological stages. However, some differences at the three physiological stages were evident in proteins of uterine fluid and in extracts of uterine tissue.Oviducal and uterine tissue fragments incubated in vitro with radioactive leucine synthesized soluble proteins and released significant amounts of newly synthesized proteins into the incubation medium. However, specific proteins whose synthesis and secretion were selectively altered under the different physiological states appeared to be minor constituents of the total secretory profile. The extensive range of newly synthesized and secreted radioactive proteins revealed by gel fluorography suggests that most proteins present in luminal fluids are directly synthesized by oviducal and uterine tissue in vivo.
