UVA and UVB Irradiation Differentially Regulate microRNA Expression in Human Primary Keratinocytes

Abstract: MicroRNA (miRNA)-mediated regulation of the cellular transcriptome is an important epigenetic mechanism for fine-tuning regulatory pathways. These include processes related to skin cancer development, progression and metastasis. However, little is known about the role of microRNA as an intermediary in the carcinogenic processes following exposure to UV-radiation. We now show that UV irradiation of human primary keratinocytes modulates the expression of several cellular miRNAs. A common set of miRNAs was influe… Show more

“…However, more work is needed to unravel the exact mechanisms that lead to PP. Recent studies have indicated that UVA and UVB irradiation differentially regulates microRNA expression in human keratinocytes as well as in the dermis . MicroRNAs are a group of small non‐coding RNAs that act as post‐transcriptional regulators by binding to complementary sequences in the 3′‐untranslated region of target genes.…”

“…The reduced DNA methylation can alter the expression of mRNAs and miRNAs. Indeed, alteration of miRNAs has been found in cells exposed to UV (119). …”

“…The keratinocytes that are located at the surface of the skin are usually affected by both UVA and UVB when a solar simulator is used. However, each wavelength band may affect the cells differently, generating distinct mechanistic pathways . Here, a narrowband light source was used to evaluate the effects of UVA without the influence of UVB, as performed by other groups .…”
