Vacuolar CAX1 and CAX3 Influence Auxin Transport in Guard Cells via Regulation of Apoplastic pH

Abstract: CATION EXCHANGERs CAX1 and CAX3 are vacuolar ion transporters involved in ion homeostasis in plants. Widely expressed in the plant, they mediate calcium transport from the cytosol to the vacuole lumen using the proton gradient across the tonoplast. Here, we report an unexpected role of CAX1 and CAX3 in regulating apoplastic pH and describe how they contribute to auxin transport using the guard cell's response as readout of hormone signaling and cross talk. We show that indole-3-acetic acid (IAA) inhibition of … Show more

“…This assumption helps to explain the observations that less Ca 2+ accumulated in cau1 and cax1/cax3 shoots ( Figure 1A; see Supplemental Figure 1A online; Cheng et al, 2005;Conn et al, 2011). Note that we do not exclude the possibility that CAX1 and CAX3 may be actively involved in the [Ca 2+ ] o signaling pathway via complex mechanisms such as reported recently (Cho et al, 2012). (A) and (B) [Ca 2+ ] o -induced stomatal closure in plants.…”

“…Color changes were documented 4 to 48 h after transfer. Apoplastic pH measurements were conducted using the infiltration/centrifugation method as previously described (Cho et al, 2012). Fluorescence of 8-hydroxypyrene-1,3,6-trisulfonic acid trisodium (Invitrogen) was determined with a Bio-Tek Synergy HT plate reader (excitation = 460 nm) and emission collected at 510 and 530 nm.…”

“…In particular, CAX1 was proposed to interfere with the positive feedback loop shown between Reactive Oxygen Species (ROS) production and Cd-induced cytosolic Ca 2C spikes, especially at low external Ca 2C supply. 4 CAX1 contribution to the cellular homeostasis, in particular as key-regulator of apoplastic Ca 2C and pH, was previously studied, 5,6 together with its capacity to shape cell wall extensibility and composition.…”

“…Cd-induced increase of AHAs transcripts in A. halleri and A. thaliana may be linked to the proton release into the cytosol in exchange of Ca 2C due to CAX1 activity. 6 Expression of genes involved in oxidative stress response (i.e., RBOHs and CAT1) was also investigated. Cd contamination enhanced RBOHD expression in A. thaliana, as previously observed by Cuypers et al (2011), 12 and not in A. halleri.…”
