Vacuolar Phosphate Transporters Contribute to Systemic Phosphate Homeostasis Vital for Reproductive Development in Arabidopsis

Abstract: Vacuolar storage of phosphate (Pi) is essential for Pi homeostasis in plants. Recent studies have identified a family of vacuolar Pi transporters, VPTs (PHT5s), responsible for vacuolar sequestration of Pi. We report here that both VPT1 and VPT3 contribute to cytosol-to-vacuole Pi partitioning. Although VPT1 plays a predominant role, VPT3 is particularly important when VPT1 is absent. Our data suggested that the vpt1 vpt3 double mutant was more defective in Pi homeostasis than the vpt1 single mutant, as indica… Show more

“…In Arabidopsis, three members of the PHT5 family are responsible for Pi storage into vacuoles ( Liu et al, 2015 , 2016 ). VPT1 (PHT5;1) is the major player in vacuolar Pi sequestration ( Liu et al, 2015 , 2016 ; Luan et al, 2019 ). Single cell profiling revealed a ubiquitous expression of VPT1 in all root cell-types, while VPT2 (PHT5;2) and VPT3 (PHT5;3) were expressed at low levels in roots of young seedlings under the normal growth conditions ( Figure 7C ).…”

“…For example, AtTPK4 is predominantly expressed in pollen, where it controls K + homeostasis and membrane voltage during pollen tube growth (Becker et al, 2004). AtCHX21 and AtCHX23 affect steps in signal reception and/or transduction that are critical to shifting the axis of polarity and directing pollen tube growth toward the ovule (Lu et al, 2011).…”

“…In addition, AtVPT1 is expressed primarily in younger tissues under normal Pi conditions, but is strongly expressed in older tissues under high-Pi conditions, suggesting that AtVPT1 functions in Pi storage in young tissues and in detoxification of high Pi in older tissues (Liu et al, 2015). Another VPT protein, AtVPT3, contributes to cytosol-to-vacuole Pi transport, but is particularly important when AtVPT1 is absent (Luan et al, 2019).…”

“…Excess Pi is toxic even at physiological concentrations of cytoplasmic Mg 2+ due to its incorporation into ATP and subsequent disruption of Mg 2+ -dependent processes. Our findings provide a conceptual framework to understand the underlying basis of Pi cytotoxicity observed in bacteria and eukaryotes (3)(4)(5)(6)(7)(8)(9)(35)(36)(37)(38). Moreover, we uncover the regulatory logic by which cells control of P assimilation.…”
