Validation of a phenol‐sulfuric acid method in a microplate format for the quantification of soluble sugars in ruminant feeds

Abstract: Soluble sugars in feeds are important for ruminant production; however, performing numerous sugar analyses within a short period is a laborious task. Here, we developed a phenol‐sulfuric acid (PSA) assay in a microplate format to quantify soluble sugars in ruminant feeds. This method is easy and quick and requires only a small quantity of harmful reagents. We found that assay measurements were not affected by the representative organic acids and sugar alcohol contained in feeds. The treatment of activated char… Show more

“…This method is carried out to determine the appropriate concentration to obtain the highest absorbance that can be read on a UV-Vis Spectrophotometer [18,19]. The test was carried out three times using several comparisons of Fucoidan concentration, 5% phenol and sulfuric acid as shown in Table 1 and Table 2.…”

“…The levan content was measured by the phenol-sulfuric acid method (PSA) with minor modifications (Kondo et al, 2021). In brief, 1 mL of natto mucus (diluted to a certain number) was mixed with 1 mL of distilled water, 1 mL of 5% phenol solution and 5 mL of concentrated sulfuric acid.…”

“…The NIR spectral data were combined with those of a colorimetric method that employs a phenol-sulfuric acid (PSA) assay in microplate format to quantify carbohydrates. 32 This method is used widely in food, algae, 33 mushrooms 34 and feed [35][36][37] because of its sensitivity and simplicity.…”
