Validation of the BIOCHIP test for the diagnosis of bullous pemphigoid, pemphigus vulgaris and pemphigus foliaceus

Abstract: BackgroundThe BIOCHIP is a novel multiplex indirect immunofluorescence technique used in the serological diagnosis of bullous pemphigoid and pemphigus. The BIOCHIP method combines the screening of autoantibodies and target antigen‐specific substrates in a single miniature incubation field.ObjectiveTo evaluate the diagnostic accuracy of the new immunofluorescence BIOCHIP multiplex tool in pemphigus and bullous pemphigoid.MethodsFor the validation of the BIOCHIP, sera from patients with BP (n = 38), PF (n = 8) a… Show more

“…Moreover, measuring autoantibodies to BP180 and BP230 in the serum is useful for diagnosing BP with high sensitivity (76.1-86.8%) and specificity (85.0-90.5%). 11,12 In this study, the serum of the four patients with available anti-BP180/BP230 data was negative for these autoantibodies, indicating a low probability for BPs.…”

“…15 The sensitivity of DSG1 transfected cells on the BIOCHIP has been reported to be between 75 and 90% for pemphigus foliaceous. 8,9 In bullous pemphigoid, the sensitivity of the BP180 mosaic on the BIOCHIP has been found to range more widely between 55.3 and 100%, [8][9][10][11][12]16,17 with a specificity between 96.5 and 100%. [8][9][10][11][12]16,17 This is compared to sensitivities of 45.1-66.7% for the BP230 mosaic on the BIO-CHIP, and a specificity of 39.0-100%.…”

[8][9][10][11][12]16,17 The lower sensitivity of the BP230 mosaic compared to the BP180 mosaic is not unexpected given that it is more common to have autoantibodies against BP180, compared to BP230 in bullous pemphigoid.…”

“…The method of serological detection of the autoantibodies largely relies on indirect immunofluorescence (IIF) and enzyme-linked immunosorbent assay (ELISA) tests. Recently, novel diagnostic multivariant assays were developed as practical screening tools for patients with suspected ABD with the aim of processing the most common autoantibodies simultaneously [39,40].…”
