Value of the Serum Alpha-L-Fucosidase Activity in the Diagnosis of Colorectal Cancer

Abstract: Objectives: The purpose of this study was to assess the value of the serum levels of α-L-fucosidase activity in the diagnosis of patients with colorectal cancer. Methods: Using a fluorometric method we analyzed the α-L-fucosidase activity in preoperative sera from 137 colorectal cancer patients and in sera from 232 donors. Results: The enzymatic activity of α-L-fucosidase was significantly lower (p < 0.001) in patients (4.8 ± 3.09 U/ml) than in donors (10.5 ± 5.46 U/ml). Using the ROC curve, the ideal cut-off … Show more

“…We have also found that in preoperative serum from CRC patients AFU activity is significantly decreased compared to healthy controls [12]. A diminished AFU activity is in agreement with the higher levels of L -fucose detected in both tumour and serum of CRC patients [13].…”

“…Serum AFU activity was measured as previously described [12], using the synthetic substrate 4-methyl-umbelliferyl-α- L -fucopyranoside (4-MU-fucoside; Sigma, St. Louis, Mo., USA). One unit of enzyme was defined as the amount of enzyme required to hydrolyse 1 nmol of substrate per minute at 37°C.…”

“…Carcinoembryonic antigen (CEA) levels in serum samples from 109 colorectal cancer patients and 72 healthy donors were analyzed by specific immunoassay using the Enzymun-Test CEA (Boehringer Mannheim, Germany) as previously described (15). CEA concentrations are expressed in ng/mL serum.…”
