Background: So far, optimization problems for diagnostics and prognostication aids have been still relevant for lung cancer as a leader in the structure of cancers. Aims and Objectives: A search for regularities of changes in the saliva enzyme activity in patients with lung cancer. Materials and Methods: In the case-control study, 859 people divided into two groups took part: Primary (lung cancer, n = 286) and control (conventionally healthy, n = 573). All the participants went through a questionnaire survey, saliva biochemical counts, and a histological verification of their diagnosis. The enzyme activity was measured by spectrophotometry. Between group differences were measured with the nonparametric test. Results: In terms of lung cancer, we observe metabolic changes described with decreased de Ritis coefficient (P ˂ 0.001), as well as the increased activity of alanine aminotransferase (P = 0.022), alkaline phosphatase (P ˂ 0.001), gamma-glutamyl transpeptidase (P = 0.043), and α-amylase (P ˂ 0.001). We have identified specifics of the change in the enzyme activity depending on a histological type and a growth form of the tumor. There is the observable increased activity of α-amylase in the saliva in lung adenocarcinoma and neuroendocrine tumors (P ˂ 0.001). It has been found that irrespective of the histological type of lung cancer in a transition from central to peripheral forms of growth; there is the observable increased activity of all the enzymes. The significantly reduced activity of aspartate aminotransferase (by 51.4% for adenocarcinoma, P = 0.032; 48.8% for squamous cell lung cancer, P = 0.042) is a distinctive feature of central lung cancer. Conclusion: The nature of the change in the enzyme activity is ambiguous and to a greater degree depends on the tumor form of growth (central/peripheral lung cancer) than the histological type (adenocarcinoma/squamous cell lung cancer).

Monotropein, the primary iridoid glycoside isolated from Morindacitrifolia, has been previously reported to possess potent antioxidant and antiosteoporotic properties. However, there is no direct evidence correlating the antiosteoporotic effect of monotropein with its observed antioxidant capacity, and the molecular mechanisms involved in mediating these processes remain unclear. Therefore, the aim of the present study was to investigate the protective effects of monotropein against oxidative stress in osteoblasts and the mechanisms involved in mediating this process. Osteoblast viability was evaluated using the MTT assay. The mitochondrial membrane potential and reactive oxygen species were detected by flow cytometry analyses. Western blotting and enzyme‑linked immunosorbent assays were performed to detect protein expression levels. A significant reduction in osteoblast viability was observed at 24 h following exposure to various concentrations (100‑1,000 µM) of H2O2 compared with untreated osteoblasts. The cytotoxic effect of H2O2 was notably reversed when osteoblasts were pretreated with 1‑10 µg/ml monotropein. Pretreatment with 1-10 µg/ml monotropein increased the mitochondrial membrane potential and reduced the generation of reactive oxygen species in osteoblasts following exposure to H2O2. In addition, the H2O2‑induced increase in apoptotic markers (caspase-3 and caspase-9) and H2O2-induced reduction in sirtuin 1 levels were significantly reversed following pretreatment of cells with monotropein. Furthermore, monotropein significantly reduced H2O2‑induced stimulation of NF‑κB expression, in addition to the expression of a number of proinflammatory mediators. These results indicate that monotropein suppresses apoptosis and the inflammatory response in H2O2‑induced osteoblasts through the activation of the mitochondrial apoptotic signaling pathway and inhibition of the NF‑κB signaling pathway.

Context:Biochemical changes occur in biological fluids and tissues of different types of malignancies. Tumor markers in serum, tissue, and other body fluids during neoplastic process are of clinical value in the management of patients with cancers. Serum alkaline phosphatase (ALP) activity is potentially a useful indicator for detection of malignancies, but its status in oral squamous cell carcinoma (OSCC) is less explored.Aims:The aim of this study is to evaluate the serum level of ALP in OSCC patients and assess its relation with the clinicopathological features.Settings and Design:A total of 175 participants (145 OSCC patients and 30 healthy controls) were included in the study. One hundred and forty-five patients with OSCC who underwent treatment at our institution were included to obtain the clinicopathological data.Materials and Methods:Fasting blood ALP activity was evaluated using ALP assessment kit and biochemistry analyzer.Statistical Analysis Used:The data were analyzed by SPSS-21 software (SPSS Statistics for Windows, Version 21.0, Armonk, NY, USA), using t-test, Mann–Whitney U, and Kruskal–Wallis tests.Results:Raised ALP was seen in 24% of OSCC patients. The mean ALP in OSCC was significantly higher than the control. ALP level in patients with advanced stage was significantly higher than with early stage. The serum ALP level in OSCC patients with bone involvement (BI) by local extension of tumor was significantly higher than without BI.Conclusion:ALP showed statistically significant differences in relation to tumor stages and BI. Hence, ALP could be useful in advanced stage disease for expressing the endurance of patient and tumor expansion. Elevated ALP in OSCC patients may indicate BI.
