Variability of mitochondrial DNA in Japanese dace, Tribolodon hakonensis (Cyprinidae).

Abstract: Intra-and inter-populational variations of Japanese dace Tribolodon hakonensis collected at three locations which were geographically distant from each other were examined with restriction fragment analysis of mitochondrial DNA (mtDNA). Twelve mtDNA types (Types I-XII) were discriminated and each type was population specific. Namely, Otsuchi Bay Population contained Types I to V, the Monobe River Population contained Types VI and VII, and Lake Biwa Population contained Types VIII to XII. The mean number of nuc… Show more

“…Shaffer and Mcknight (1996) revealed that within-locality variation was 0.1% in Ambystoma velasci and A. tigrinum nebulosum. Hanzawa et al (1987) estimated that the expected intrapopulational variability was 0.07~0.23% in Japanese dace, Tribolodon hakonensis. Avise and Lansman (1983) reported that the intrapopulational variability of pocket gophers was less than 1%.…”

“…The individuals used for the analyses did not contain hybrids, because they were exactly identified by differential alleles substituted among species (Hanzawa and Taniguchi, 1982;Hanzawa et al, 1987Hanzawa et al, , 1988Sakai, 1995;Kartavtsev et al, 2002) and morphological difference of cephalic lateral-line systems (Nakamura, 1963(Nakamura, , 1969Kurawaka,1977). Total DNA was purified from fresh tissue of skeletal muscle or liver according to the method described by Asahida et al (1996).…”

“…The data set obtained was converted into binary matrices with respect to the restriction sites. Haplotype (h) and nucleotide (π) diversity, as well as the level of nucleotide divergence (p) within and among the species were calculated using the equations of Nei [15] and Nei and Tajima [16] as implemented in the REAP software package [24] for the restriction endonuclease digestion data. Phyloge netic analysis was carried out using the Bayesian Infer ence (BI) in the MrBayes v3.12 software program [25].…”

“…Comparison of karyological and genetic data obtained will make it possible to clarify the taxo nomic status of T. hakonensis, as well as to reveal the differences and similarities between the two forms within the species of interest. Intraspecific variation and phylogenetic relation ships of Tribolodon species are well studied in some Japanese populations, which were examined using allozymes and mtDNA variation [13][14][15], as well as sequence data for the 16S rRNA, cyt b, and D loop fragments [13,16]. At the same time, only few studies dealt with the genus Tribolodon in the Russian part of the range using the same approaches [11,17].…”
