Variable Number Tandem Repeat Profiling of Mycobacterium ulcerans Reveals New Genotypes in Buruli Ulcer Endemic Communities in Ghana and Côte d’Ivoire

Abstract: Buruli ulcer (BU), a necrotic skin disease caused by Mycobacterium ulcerans, is mainly prevalent in West Africa, but cases have also been reported in other tropical parts of the world. It is the second most common mycobacterial disease after tuberculosis in Ghana and Côte d’Ivoire. Heterogeneity among M. ulcerans from different geographical locations has not been clearly elucidated, and some studies seem to suggest genetic differences between M. ulcerans in humans and in the environment. This study aimed at id… Show more

“…Although the in-house isolates were MU strains from Africa lineage 1 and shared pseudogenes with the African isolates, their pseudogenome pattern was less similar to the African MU strains, especially PA38. Multiple loci VNTR genotyping shows that Z007, Z039, and PA38 have the common African profiles C and D [ 21 ]. PA38 further clustered with a strain from Benin pointing to the possible origin of the PA38 strain.…”

“…Buruli ulcer and lymphatic filariasis case numbers followed the same trend as leprosy in Cameroon and Côte d'Ivoire, and tungiasis was only recorded in Cameroon. Buruli ulcer and lymphatic filariasis are known to be endemic in Cameroon, Côte d'Ivoire and Ghana (11)(12)(13)(19)(20)(21) suggesting that the diseases may have similar risk factors, and that tungiasis may be more common in Cameroon due to specific environmental factors.…”

“…In Cameroon, Côte d'Ivoire, and Ghana, several skin-NTDs are known to be endemic including yaws (7)(8)(9)(10), Buruli ulcer (11)(12)(13), leishmaniasis (14)(15)(16), leprosy (10,17,18), lymphatic filariasis (19)(20)(21), mycetoma (22)(23)(24), onchocerciasis (25)(26)(27), and scabies (10,(28)(29)(30). Alongside these conditions, a number of more common skin conditions including superficial fungal and bacterial infections cause a considerable burden of disease (28,(31)(32)(33).…”
