Abstract. During August 2009, measurements of bacterial abundance and nucleic acid content were made along with production and respiration in coastal waters of the Beaufort Sea (Arctic Ocean), an area influenced by the Mackenzie River inflow. The main purpose was to evaluate bacterial organic carbon processing with respect to local sources, mainly primary production and river inputs. Bacterial production and abundance generally decreased from river to offshore waters and from surface to deep waters. In contrast, the percentage of high nucleic acid bacteria was higher in deep waters rather than in surface or river waters. Statistical analyses indicated that bacterial production was primarily controlled by temperature and the availability of labile organic matter, as indicated by total dissolved amino acid concentrations. Direct comparisons of bacterial carbon demand and primary production indicated net heterotrophy was common in shelf waters. Net autotrophy was observed at stations in the Mackenzie River plume, suggesting that the carbon fixed in plume waters helped fuel net heterotrophy in the Beaufort Sea margin.

Abstract. Bacteria with low nucleic acid content (LNA) and high nucleic acid content (HNA) are widely distributed in aquatic environments. Most of the current understanding of these two subgroups is derived from studies in marine environments. In comparison, information on the spatial distribution of these two subgroups in freshwater environments is very limited. The present study analysed the biogeographical pattern of those two groups on a large-river scale (i.e. the Songhua River catchment, .1000 km). The results showed that the concentrations of LNA and HNA bacteria were distributed over a wide range from 5.45 Â 10 4 to 4.43 Â 10 6 cells mL À1 , and from 1.35 Â 10 5 to 4.37 Â 10 6 cells mL À1 respectively. The two groups have almost equal proportions in the Songhua River, with the average contribution of LNA bacteria reaching 47.0%. In comparison, the abundance of LNA bacteria in the mainstream was significantly higher than in the tributaries. The cytometric expressions (green fluorescence and side scatter) within LNA and HNA were strongly covaried, which implies that these two subgroups are intrinsically linked. Multivariate redundancy analysis indicated that both the abundance and cytometric characteristics of co-occurring LNA and HNA bacteria were regulated differently in the Songhua River. This suggests that LNA and HNA bacteria play different ecological roles in river ecosystems.

Sea ice, a characteristic feature of polar waters, is home to diverse microbial communities. Sea-ice picoeukaryotes (unicellular eukaryotes with cell size o3 lm) have received little attention compared with diatoms that dominate the spring bloom in Arctic first-year sea ice. Here, we investigated the abundance of all picoeukaryotes, and of 11 groups (chlorophytes, cryptophytes, bolidophytes, haptophytes, Pavlovaphyceae, Phaeocystis spp., pedinellales, stramenopiles groups MAST-1, MAST-2 and MAST-6 and Syndiniales Group II) at 13 first-year sea-ice stations localized in Barrow Strait and in the vicinity of Cornwallis Island, Canadian Arctic Archipelago. We applied Catalyzed Reporter Deposition-Fluorescence In Situ Hybridization to identify selected groups at a single cell level. Pavlovaphyceae and stramenopiles from groups MAST-2 and MAST-6 were for the first time reported from sea ice. Total numbers of picoeukaryotes were significantly higher in the vicinity of Cornwallis Island than in Barrow Strait. Similar trend was observed for all the groups except for haptophytes. Chlorophytes and cryptophytes were the dominant plastidic, and MAST-2 most numerous aplastidic of all the groups investigated. Numbers of total picoeukaryotes, chlorophytes and MAST-2 stramenopiles were positively correlated with the thickness of snow cover. All studied algal and MAST groups fed on bacteria. Presence of picoeukaryotes from various trophic groups (mixotrophs, phagotrophic and parasitic heterotrophs) indicates the diverse ecological roles picoeukaryotes have in sea ice. Yet, 450% of total sea-ice picoeukaryote cells remained unidentified, highlighting the need for further study of functional and phylogenetic sea-ice diversity, to elucidate the risks posed by ongoing Arctic changes.
